Intrasplenic Autotransplantation of Hepatocytes in Dogs
Authors
Abstract:
Objective- The liver has a unique location in the body for performing multiple functions such as,metabolism, detoxification and synthesis. There are limitation for liver transplantation due to shortage ofavailable donors and requires for sophisticated technology and support surgical teams. The purpose of thisstudy was to investigate the hepatocyte transplantation into the spleen of animal model such as dog toestabli sh basic condition for further studies.Design- In vivo experimental Study.Animals- Five Iranian indigenous mixed breed dogs of both sexes, 15/8±3 months old and 19.6±8.2weighting were used in this sudy.Procedures- Ventral midline celiotomy was performed under general aneasthesia and liver sampls wereobtained using suction technique by a 50cc syring and 14 gauge needle . Then obtained samples wereinjected into the same dog 's spleen directly and also via splenic artery, the celiotomy incision sutured andthe animal left to recover. On the 16th postoperative day, under general anaesthesia, the gross apearance ofspleen were recorded and photographed and for histpathological evaluation, partial splenectomy wasperformed. The prepared sections were stained with Hemotoxylin Eosin [H& E] and Periodic Acid Schiff[PAS] and studied under light microscoy.Results- The gross evaluations ofthe removed spleen showed a whitish area at the injection site in all ofthe 5 transplanted dogs, spleen vessels had been dilated and enlarged in diameter. Microscopically thegroups of hepatocytes in the center of lymphoid follicles in the white pulp were noted. Conclusion and Clinical Relevance- Hepatocyte transplantation could be useful for both supporting anacute liver failure and for surviving as a bridge to liver tansplantation in terminal liver failure.

The role of corticosterone in the metabolic recovery after intrasplenic adrenal autotransplantation in rats.
Transplantation of adrenal cortical tissue may represent an alternative treatment to reestablish glucocorticoid secretion in adrenal insufficiency. In the present work, performed in adrenalectomized rats and adrenalectomized rats with a complete autotransplanted adrenal into the spleen, several hormones and biochemical parameters were measured and compared to control animals, in order to examin...

In virgin mice of strain A, anterior hypophyseal transplants increased the incidence of mammary cancers; in castrate males bearing hypophyseal and ovarian grafts, the number of mammary can eers exceeded that noted after transplantation of ovaries alone. These findings were interpreted as owing to the release of gonadotrophins by the hy pophyseal transplants, which stimulate the output of estrog...

OBJECTIVES The aim of this study was to investigate the effect of basic fibroblast growth factor (FGF-2) on root resorption after delayed autotransplantation in dogs. STUDY DESIGN Mandibular second and third premolars of beagle dogs were extracted to create sites for autotransplantation. After 2 months, in the experimental sites the first and fourth mandibular premolars were extracted and air...
full textExtrinsic reinnervation of the intestine after small bowel autotransplantation in dogs.
I MMUNOHISTOCHEMICAL examination of two failed human small intestinal grafts showed no evidence of extrinsic reinnervation for up to 22 or 8 months whereas intrinsic neural innervation showed only a slight decrease from normal intestine. The mechanisms responsible for this lack of neural input are not known. The purpose of this study was to examine extrinsic reinnervation in canine small bowel ...
full textAmelioration of intestinal dysmotility and stasis by octreotide early after small-bowel autotransplantation in dogs.
BACKGROUND Intestinal dysmotility and stasis after intestinal transplantation are considered to promote bacterial overgrowth and translocation. Two prokinetic agents, KW5139 (13-leu-motilin) and the somatostatin analogue octreotide acetate, were studied to determine whether they can ameliorate intestinal dysmotility during the early postoperative period. MATERIALS AND METHODS Motility was rec...
