In vitro Susceptibility Study of Salmonella typhimurium Mutant to Stress Factors
Authors
Abstract:
ABSTRACT Background and Objective: In this study, we compared the susceptibility of Salmonella typhimurium phoP mutant and its parent to stress conditions that the pathogen may encounter in a host. Methods: For this purpose, we used the phoP deletion mutant constructed in our previous study. In order to test the in vitro susceptibility of the mutant to stress factors, the effect of acidic pH, heat, bile salts and polymyxin on growth of the mutant was examined. Then, minimum inhibitory concentration and minimum bactericidal concentration of bile salts and polymyxin were determined. Salmonella typhimurium 14028 was used as the parent strain. Results: The mutant was highly susceptible to bile salts and polymyxin in comparison with the parent strain, but no difference was observed in their susceptibility to acid and heat. Conclusion: This study confirms the role of the phoP in resistance of Salmonella to polymyxin and bile salts. Mutation in the phoP leads to susceptibility of the mutant to bile and cationic antimicrobial peptides. Keywords: In vitro, Mutant, Salmonella typhimurium, Disease Susceptibility.

Virulence of a Salmonella typhimurium OmpD mutant.
An ompD mutation caused by a Tn10 insertion was transduced into Salmonella typhimurium SL1344 and UK-1. The adherence and invasion capabilities of the resultant ompD mutants were examined by tissue culture analysis. The virulence of the S. typhimurium ompD mutants was ascertained by a 50% lethal dose (LD50) study and by determining colonization ability with BALB/c mice. We found no statisticall...
full textOsmotic-sensitive mutant of Salmonella typhimurium.
A strain (DA82) having peculiar osmotic properties was isolated in Salmonella typhimurium. The mutant shows increased elasticity of its cell wall and makes spherical instead of elongated cells, regardless of the osmolality of the medium. The strain withstands dilution in distilled water without disruption or death and grows normally in 0.1 molal NaCl broth (240 milliosmol), but it dies exponent...

ses. Enzymatic determinations demonstrated, however, that the enzymes of the glycolytic pathway (up to the formation of triose phosphates) were present and active at 25 and 37 C. At 42 C, the mutant did not grow with any of the carbon sources used. At both 37 and 42 C, the mutant grew perfectly well with hexoses if yeast extract was present. The metabolite required for growth was thiamine or, s...
full textIsolation and Detection of Virulence factors of Salmonella typhimurium and Salmonella enteritidis
Plasmids play an important role in the virulence and antibiotic resistance of Salmonella. Salmonella carry plasmids having different size and number. Plasmid profile analysis of single isolates of Salmonella typhimurium and Salmonella enteritidis revealed the presence of 4- plasmid profiles. Salmonella serovars produce several Type III secretions including Sop-E and Sop –B. Soap E is detected b...
full textIn vitro polymerization of flagellin excreted by a short-flagellum Salmonella typhimurium mutant.
The culture medium of a short-flagellum mutant of Salmonella typhimurium contained a large amount of mutant flagellin and a small amount of strong inhibitors of flagellin polymerization. After being freed of the inhibitors, the mutant flagellin could be polymerized in vitro, although under nonphysiological conditions.
