IN VITRO STUDY OF AN ENDOGENOUS IMMUNOSUPPRESSOR FACTOR DERIVED FROM HUMAN OR BOVINE SERUM

Abstract:

The effects of the human and bovine LSF (derived from sera) as well as their purified fractions were studied on murine lymphocytes reactions indicated by blast transformation (BT) assay, mixed lymphocyte culture (MLC) and IgG synthesis. The results indicated that bovine lipid suppressor factor (LSF) has significant immunosuppressive activity on lymphocytes proliferation both in BT and MLC assays. However, human LSF showed suppressive activity on lymphocyte proliferations only in MLC but not in BT assays. Concerning the effects of partially purified bovine LSF molecules on humoral immune response, we found that band #3 at 1:s dilution strongly suppresses IgG synthesis of murine spleenocytes in response to pokeweed mitogen stimulation. Band #1 and band #3 are agents with cellular immunosuppressor activity. However, band #2 is not as suppressive as the other bands in BT, MLC, and IgG assays. It seems that the mixture of all bands together may have synergistic activity in the in vitro studies. This idea has been concluded from the results in BT and MLC assays which are presented in histograms #1 and #2. Before concluding that we have found an alternative immunotherapeutic agent to cyclosporine A, we need to study the molecular structure of these compounds as well as in vivo reactions of bovine LSF in skin transplantation. Since purified human LSF and its three bands did not indicate as much cellular immunosuppressive activity as bovine LSF, the study of their effects was not followed up in detail like bovine LSF in this project

Labeling of Human Serum Albumin with Stable Isotope of Bromine; an in Vitro Study

Background: Possibility to trace-label albumin with isotopes results in information concerning its synthesis, breakdown, and distribution in the intra and extra cellular spaces. The iodination of albumin is a widespread procedure used in scientific studies. Bromine not only is more reactive and less expensive than iodine, but bonds more easily with many elements. Therefore, it could be a suitab...

Human Adipose-Derived Stem/Stromal Cells from Children or Adults?

Discovering a cell type with the highest efficiency and the least side-effects is one of the priorities of scientists active in this field. Of all the cells, mesenchymal stem cells have been and still are of interest to the researchers for use in pre-clinical and clinical studies due to being easily accessible, pluripotency, secretion of numerous growth factors, low risk of immunological reject...

Differentiation of adipose-derived stem cells into Schwann-like cells: fetal bovine serum or human serum?

Access to autologous Schwann cells is limited due to lack of donor site and its difficult isolation and culture. Therefore, one of the possible ways to obtain to Schwann cells is to differentiate mesenchymal stem cells into glial pathway using various materials and protocols. The aim of this study was to compare the effects of fetal bovine serum and human serum on Schwann cell differentiation o...

assessment of surface markers derived from human periodontal ligament stem cells: an in vitro study

objectives: periodontal tissue regeneration for treatment of periodontal disease has not yet been mastered in tissue engineering. stem cells, scaffold, and growth factors are the three main basic components of tissue engineering. periodontal ligament (pdl) contains stem cells; however, the number, potency and features of these cells have not yet been understood. this study aimed to isolate and ...

Differentiation of Mesenchymal Stem Cells Derived From Human Adipose Tissue into Cholinergic-like Cells: In Vitro Study

Introduction: Cholinergic-associated diseases currently constitute a significant cause of neurological and neurodegenerative disabilities.  As the drugs are not efficient in improving the suffered tissues, stem cell treatment is considered an effective strategy for substituting the lost cells. Methods: In the current study, we set out to investigate the differentiation properties of human adip...
