Immunization of Arabian sheep with whole gut homogenate of Haemonchus contortus
Authors
Abstract:
Ten female lambs of 7-month-old were divided into two equal groups and raised under a helminth-free conditions. Animals in group 1 were immunized two times by whole gut homogenate (WGH) of Haemonchus contortus emulsified in Freund’s adjuvant. In group 2 (control), animals were injected by phosphate buffered saline emulsified in the same adjuvant. Animals were challenged by 10000 third-stage larvae (L3) of Haemonchus contortus on day 33 after the first immunization and then humanely killed on day 42. Animals were tested for serum antibody and eggs per gram of faeces (EPG) throughout the study and nematodes in their abomasom were counted after necropsy. The results indicated that animals immunized with WGH showed a higher level of serum antibodies. A significant difference was observed in mean optical density of sera in ELISA between the two groups (P

full textImmunological responses of sheep to Haemonchus contortus.
Infections with Haemonchus contortus are a major constraint on ruminant health world-wide. Young lambs are very sensitive to Haemonchus infection. Older lambs and sheep acquire immunity after a continuous or seasonal exposure to the parasite. The mechanisms underlying immunity are still not completely understood. Antibodies, in particular local IgA and IgE, certainly play a role. The role of Ig...

full textImmunodiagnosis of Haemonchus contortus infection in sheep by indirect enzyme linked immunosorbent assay (ELISA)
Indirect plate enzyme-linked immunosorbent assay was standardized and evaluated for its effectivenessin immunodiagnosis of haemonchosis in experimental and clinical cases in sheep by using somatic wholeadult antigen of H. contortus. Plate ELISA was standardized using 5 μg/well antigen concentration with1:100 and 1:1000 of sera and conjugate dilution. Indirect plate ELISA was able to demonstrate...
full textEvaluation of crude larval protein and recombinant somatic protein 26/23 (rHcp26/23) immunization against Haemonchus contortus in sheep
AIM The aim of this study was to evaluate the potential possibility of crude larval and recombinant (rHcp26/23) antigens of Haemonchus contortus for immunization to control sheep hemonchosis. MATERIALS AND METHODS A total of 21 lambs were divided into five groups. Lambs were immunized with larval and recombinant (rHcp26/23) proteins at day 0 and day 14 and after that challenged with 5000 infe...
full textImmunization against Lamb Haemonchosis with a Recombinant Somatic Antigen of Haemonchus contortus (rHcp26/23)
Haemonchosis, caused by the abomasal nematode Haemonchus contortus, is a common parasitic disease of sheep. Our previous results showed that a soluble fraction from adult stages of the nematode (p26/23) induced partial protection against challenge. Recombinant DNA technology was applied to obtain a synthetic protein (rHcp26/23). Immunological assays (ELISA, Western blotting, and immunolocalizat...
