Abstract:

Background To assess embryos derived by the transfer of meiosis-II chromosomes (M-II-t) from aged mice oocytes into ooplasms from younger mice to overcome the problem of age-related decline in female fertility. MaterialsAndMethods The developmental capacity karyotype and ultrastructure of reconstructed oocytes derived from meiosis-II chromosome transplantation from aged mice into the ooplasms of young mice by piezo-micromanipulation were assessed. Results The survival rate of enucleated young oocytes was 54% and the percent of fertilized reconstructed oocytes was 23%. The rate of embryo development to the two-cell stage after cultivation was 40%. Since 82.4% of the analyzed embryos derived from reconstructed oocytes had condensed nuclei it was not possible to analyze their chromosomal integrity. However 17.6% of analyzable reconstructed old oocyte derived embryos (old-ODEs) had normal diploid sets of chromosomes. Major structural differences were not observed between young old and M-II-t derived two-cell embryos. Conclusion Our findings suggested that ooplasms from younger mice may overcome ageassociated problems in older mice.

the arrangement of microtubules in embryos derived from mice young, old and reconstructed oocytes

objective: to study the structure and distribution of microtubules in embryos derived from young, old and reconstructed oocytes. materials and methods: embryos obtained from old (50 embryos), young (50 embryos) and reconstructed oocytes (10 embryos) were studied by immunocytochemistry. the microtubule structures of the embryos were studied by using fluroscent microscopy with fitc-pi filter and ...

Cytogenetic analysis and developmental assessment of mouse embryos derived from in vitro fertilization of oocytes reconstructed by meiosis-II chromosome transplantation.

An electrofusion methodology for transferring meiosis-II chromosomes (M-II-t) has not been completely established. The present study compared the use of two temperatures (fusion at 37 C for Group A and 25 C for Group B) during an electrofusion procedure for mouse oocyte M-II-t and investigated the cytogenetic normality and developmental competence of embryos derived from in vitro fertilization ...

P-118: Developmental Competence of Ovine Embryos Derived from Frozen Oocytes Co-Cultured with Embryos Derived from Fresh Oocytes

Background: Currently, a limited number of studies have been done on the vitrification of small ruminant oocytes, especially in sheep, where poor developmental rates are obtained following immature and mature oocyte vitrification. Poor ovine oocyte cryopreservation could attribute to the damage to enzymes which has role in metabolic pathways. Therefore, production of cytokines and growth factor...
