Abstract:

Background: Acanthamoeba is a free-living and opportunistic amoeba that the potential of this parasite to convert to a cyst, making its treatment difficult. In this study, we investigated the effect of microwave radiation on Acanthamoeba cysts in vitro. Materials and Methods: Acanthamoeba cysts were reproduced in the culture medium. We divided 16 test tubes containing cysts into two groups. The contents of 8 tubes were irradiated with continuous microwave in the time range of 0-120 s, and the next 8 tubes were irradiated with repetitive microwave in the range of 0-360 s. The mortality of cysts at the end of irradiation was recorded and compared with control. Results: Continuous and repetitive irradiation of microwave resulted in mortality of 100 (%) of Acanthamoeba cysts during 120 and 360 seconds, respectively. In continuous mode, parasite mortality in all groups was significantly different from the control group (P<0.0001).  But in the repetition pattern only in groups that had been irradiated for more than 60 seconds, parasite mortality was significantly different from the control group (P<0.0001). Conclusion: The change of radiation mode from continuous to repetitive, reduces the thermal effects of the microwaves but does not affect the mortality rate of Acanthamoeba cysts. Therefore, probably the impact of microwaves was not only caused by the increase in ambient heat but also its another property is involved in the death of Acanthamoeba.

Effect of steroids on Acanthamoeba cysts and trophozoites.

PURPOSE Topical steroids are frequently used to control corneal inflammation and uveitis or is administered after surgery, to prevent corneal graft rejection. This study was undertaken to determine whether steroids could affect the pathogenicity of Acanthamoeba castellanii. METHODS The effect of dexamethasone phosphate on excystment, proliferation, and encystment of trophozoites and cysts of ...

Effects of Aqueous and Ethanolic Extracts of Myrtus Communis Leaves on Trophozoites and Cysts of Acanthamoeba: An In Vitro Study

Background and Aims: Acanthamoeba is a ubiquitous amphizoic organism which can cause lethal diseases such as granulomatous amoebic encephalitis and unfortunately, the infection has now increased in the world. The aim here was to evaluate in vitro anti-Acanthamoeba properties of crude aqueous and ethanolic extracts of Myrtus communis. Materials and Methods: In this experimental research, a clin...

Effect of caspofungin on trophozoites and cysts of three species of Acanthamoeba.

OBJECTIVES Amoebic keratitis is difficult to treat, without total efficacy in some patients because of cysts that are less susceptible than trophozoites to the usual treatments. We investigated here the in vitro effectiveness of caspofungin, a new antifungal, against three species of Acanthamoeba. METHODS Trophozoites and cysts of Acanthamoeba castellanii, Acanthamoeba culbertsoni and Acantha...

Antimicrobial action of biguanides on the viability of Acanthamoeba cysts and assessment of cell toxicity.

PURPOSE To assess dose- and concentration-dependent rates of biguanides on the viability of Acanthamoeba cysts isolated from severe ulcerative keratitis, and to correlate cysticidal activites with cytotoxic profiles in corneal and endothelial cells. METHODS Cysticidal activities of polyhexamethylene biguanide and chlorhexidine digluconate were evaluated in the Acanthamoeba castellanii strain ...

Lethal Effects of Helianthemum lippii (L.) on Acanthamoeba castellanii Cysts in Vitro

Acanthamoeba spp. commonly cause Acanthamoeba keratitis which is typically associated with the wear of contact lenses. Therefore, finding an economic, efficient, and safe therapy of natural origin is of outmost importance. This study examined the in vitro lethal potential of ethyl acetate and methanol extracts of Helianthemum lippii (L.) (sun roses) against Acanthamoeba castellanii cysts isolat...
