Abstract:

The objective of this study was to specify the effects of centrifugation and different surface of soybean lecithin-based extender on post-thaw Sperm quality of Ghezel ram semen. Semen samples were gathered from 5 mature Ghezel ram two/week for 3 weeks. After initial evaluation, the approved semen samples were pooled together and split into equal parts in Falcon tubes. Three samples were combined with Tris buffer at 30 ˚C and then centrifuged to remove the seminal plasma. After centrifugation and removal of the supernatant, samples were diluted with Tris-citrate-fructose extender + different concentrations of soybean lecithin (1%, 1.5% and 2% weight/volume) with 7% glycerol; and residual one samples, which were not centrifuged, were diluted with the (1.5% weight/volume) soybean lecithin. Samples then cooled to 5 ˚C and frozen in 0.25 mL French straws and straws were thawed in a 37 ˚C water bath. The results of different concentrations of soybean lecithin showed that total motility (TM) and motion parameters of average path velocity (VAP), straight-line velocity (VSL), curvilinear velocity (VCL) and lateral head displacement (ALH) and and the percentage of spermatozoa with intact acrosomes were significantly higher in soybean lecithin (SL) 1.5% compared to other semen extenders (P

In Vitro Comparison of Soybean Lecithin Based-Extender with Commercially Available Extender for Ram Semen Cryopreservation

Background Egg yolk is the main cryoprotectant agent in semen freezing extenders which is used in order to protect spermatozoa against cold shock. However elimination of animal bioproducts from the cryopreservation protocol is becoming mandatory. Therefore the aim of this study is to compare a previously studied homemade soya bean lecithin based extender with a commercially available extender f...

P-137: Improvement in Post-Thawed Ram Sperm Quality by Adding Cysteine in Soybean Lecithin Based Extender

Background: Ram sperm have a higher polyunsaturated fatty acids and a lower cholesterol/phospholipid ratio than other species which renders the sperm much more vulnerable to oxidative damage caused by reactive oxygen species (ROS). Oxidative stress can affects the integrity of cells at both the structural and sub structural levels. Therefore, a suitable antioxidant may prevent cryo-injuries and...

Supplemental Glutathione Improves Post-Thaw Quality of Holstein Bulls Sperm in a Nanomicelle based Extender

Adding antioxidants to semen extenders could reduce post-thaw sperm damages. Glutathione is an important intracellular antioxidant that has free radicals scavenging properties and multiple effects on DNA and protein synthesis. In the present study, effects different levels of reduced glutathione (GSH) containing 0 (control), 1.0, 2.5, or 5.0 mM in a nanomicelle-based extender on post-thaw quali...

effect of osmolarity and glycerol different levels in soybean lecithin-based extender on bovine sperm quality after cryopreservation

the effect of osmolarity and glycerol different levels in soybean lecithin-based extender on the bull sperm quality after cryopreservation was examined using six holstein bulls in a 2 × 3 factorial trial based on completely randomized design, with three levels of osmolarity (250, 300 and 350 mosml) and two levels of glycerol (5 and 7 %) .in general, semen samples were collected 36 times (from e...
