Effective in vitro seed germination and direct regeneration from cotyledonary leaf explants of Nitraria schoberi
Authors
Abstract:
Nitraria schoberi (Ghar-e-Daghin in Persian) from Zygophyllaceae (Nitrariaceae) family is considered as a wonderful exquisite plant in terms of its economical, medicinal, animal feed, significant values and also its unique characteristics of being tolerant to the soil salinity, drought and erosion. On the other hand, this plant is challenging with severe problems such as seed dormancy, plant growth inhibitors and also hard seed coat which lead to retard germination and subsequently to reduce the chance of life and reproduction. The present study was conducted for the first time to cope with its seed germination obstacles and to induce plantlet regeneration in N. schoberi through tissue culture method. Effect of cotyledonary leaf and hypocotyl explants and various concentrations of plant growth regulators were studied on shoot and root induction and further development. Based on the obtained results, culturing seeds after removing its seed coat effectively improved germination rate (80%) within a week. Whereas, germination of seeds with seed coats required a longer time (4 weeks) and showed a sharp decline in percentage (10%). At the next step, culturing cotyledonary leaf explants on shoot induction media including MS medium supplemented with BAP (2.0 mgL-1) and IBA (0.5 mgL-1) gave the highest shoot regeneration rate (93.33%), with average shoot number per explant (3.67) and shoot length (4.6 cm) after 60 days. Hypocotyl explants showed no response to shoot induction treatments even after 60 days. Regenerated shoots were rooted on the MS medium supplemented with IAA (1.0 mgL-1) alone or in combination with IBA (2.0 mgL-1). Finally, all regenerated plantlets were acclimatized and transferred to the green house successfully with 70% survival rate.

Considering great medicinal value of Hyssopus officinalis L. and possibility of its mass production through in vitro culture, two individual experiments was conducted. Effect of various concentrations (0, 2.2, 4.4 and 11 µmol) of TDZ and BAP in combination with 1 µmol of IAA on direct regeneration from nodal explants were assessed. Significant difference between treatments was observed (P≤ 0.01...

The present study was undertaken to investigate the possibility for developing Efficient Plant regeneration protocol in Vigno mungo L. For this, the effect of Cytokinens on in vitro regeneration from Cotyledonary explants was studied in Black gram (Vigno mungo L.) Hepper Var Vamban-1. The cytokinins used were 6-benzylaminopurine (BAP), Thidiazuron (TDZ), and kinetin (Kn). The regenerated shoots...

The present work aimed at regenerating plants of Eucalyptus camaldulensis from the cotyledonary explants and describing the anatomy of the tissues during callogenesis and organogenesis processes, in order to determine the origin of the buds. The cotyledonary leaves of E. camaldulensis were cultured in Murashige and Skoog (MS), WPM and JADS media supplemented with 2.7 μM NAA and 4.44 μM BAP. The...

Direct regenerations by using mature cotyledonary node as a explants has been shown to be time-saving and convenient strategy for micropropagation of soybean. So we have evaluated regeneration protocol through single shoot using cotyledonary node as a rapid and efficient protocol for two soybean cultivars and one mutant line. Cotyledonary nodes explants obtained from 7-days-old in vitro</em...

Different explants of Crocus sativus L. such as corm, style, ovary, petal, anther and leaf were cultured on LS and MS media supplemented with NAA and BAP or 2,4-D plus BAP at different combinations. Shoot and corm induction occurred in addition to the formation of other organs. Efficient plantlet regeneration with strong roots occurred at 1 mg 1-I of both NAA and BAP. An increase in 2,4-D l...
