Abstract:

Objective(s): In the current work, poly D, L lactide-co-glycolide (PLGA) particles were applied for a viral vaccine for the delivery of antigens in cytosolic pathway by increasing the antigen presentation to T-lymphocytes. HPV-E7 protein with PLGA particles has been reported as a potent adjuvant for HPV vaccine by encapsulating protein into the PLGA particles. Polysaccharide from Pleurotus sajor-caju was also applied as a potent immunomodulator. Materials and Methods: HPV-E7 protein and Pleurotus sajor-caju polysaccharides (PSC) were encapsulated into PLGA nanoparticles. This combination comprised a strategy to induce helper and cytotoxic T-lymphocytes (CTL) expansion.  Mice antibodies and T-lymphocyte expansion were investigated in comparison between encapsulated E7 protein into PLGA nanoparticles (E7PLGA) and E7 protein with PSC encapsulated into PLGA nanoparticles (PSC-E7PLGA). Results: The results showed that E7PLGA and PSC-E7PLGA could induce antibody response to HPV. The PSC-E7PLGA could increase the level of viral antigen-specific IgG antibodies. The cellular immune responses are also significantly enhanced by expansion of helper T-lymphocytes and CTL. PSC-E7PLGA was shown to be significantly higher in immunomodulating activity than E7PLGA. Conclusion: Thus, the encapsulation of polysaccharide and HPV-E7 antigen into PLGA nanoparticles is a strategy for development of HPV vaccine.

Antitumoral activity of L-ascorbic acid-poly- D,L-(lactide-co-glycolide) nanoparticles containing violacein

It has been demonstrated that tumoral cells have a higher uptake of ascorbic acid compared to normal cells. This differential characteristic can be used as a way to improve the specificity of antitumoral compounds if combined with polymeric drug delivery systems. The aim of this study was to prepare, characterize and evaluate the antitumoral activity of poly- D,L-(lactide-co-glycolide) 50:50 lo...

Multifunctional hydroxyapatite and poly(D,L-lactide-co-glycolide) nanoparticles for the local delivery of cholecalciferol.

Cholecalciferol, vitamin D3, plays an important role in bonemetabolism by regulating extracellular levels of calcium. Presented here is a study on the effects of the local delivery of cholecalciferol (D3) using nanoparticulate carriers composed of hydroxyapatite (HAp) and poly(D,L-lactide-co-glycolide) (PLGA). Multifunctional nanoparticulate HAp-based powders were prepared for the purpose of: (...

Evaluation of the antibacterial activity of poly-(D,L-lactide-co-glycolide) nanoparticles containing violacein

Since violacein—an antibiotic, antiviral, and antiparasitic compound—exhibits poor solubility in water, polymeric poly-(D,L-lactide-co-glycolide) nanoparticles containing this compound improved its solubility and biological activity. The nanoparticles were prepared by the nanoprecipitation method and characterized in terms of average diameter, zeta potential, drug loading, polymer recovery, in ...

Angiogenic and Osteogenic Coupling Effects of Deferoxamine-Loaded Poly(lactide-co-glycolide)-Poly(ethylene glycol)-Poly(lactide-co-glycolide) Nanoparticles

Angiogenesis and osteogenesis coupling processes are essential for bone regeneration, and human bone marrow stromal cells (hBMSCs) along with endothelial cells (ECs) are crucial participants. Deferoxamine (DFO), a hypoxia-mimetic agent, could activate the hypoxia-inducible factor (HIF)-1α signaling pathway and trigger angiogenic and osteogenic effects in these cells. However, the lifetime of DF...

Preparation and Characterization of Poly(D,L-Lactide-co-Glycolide) Nanoparticles Containing Ascorbic Acid

This paper is covering new, simplistic method of obtaining the system for controlled delivery of the ascorbic acid. Copolymer poly (D,L-lactide-co-glycolide) (DLPLG) nanoparticles are produced using physical method with solvent/nonsolvent systems where obtained solutions were centrifuged. The encapsulation of the ascorbic acid in the polymer matrix is performed by homogenization of water and or...

Poly(D, L-lactide-co-glycolide)/Poly(ethylenimine) Blend Matrix System for pH Sensitive Drug Delivery

Poly(d,l-lactide-co-glycolide) (PLGA) and poly(ethylenimine) (PEI) were blended and found to form a homogeneous pH sensitive matrix for drug release. Differential scanning calorimetry (DSC) studies of the PLGA/PEI blends showed a single glass transition temperature at all compositions. Fourier transform infrared spectroscopy (FTIR) demonstrated that the PLGA carbonyl peak at 1760 cm 1 shifted t...
