Abstract:

Introduction: Failure of human body tissue and organs is believed to be one of the most important health problems all over the world. The great challenge for tissue engineers is to optimize suitable systems to separate, proliferate and differentiate the cells so that they can set out to create tissue by a harmonic 3-D growth. Therefore, the tissue engineers must provide an environment like the living cells environment. Methods: In this study, to increase efficiency, the nano fiber structure of poly lactic acid modified with glycosaminoglycan. Then the mesenchymal cells of bone marrow cultured on this 3-D structure with differentiated media culture of TGF-ß .To investigate differentiation, the genes expressed in cartilages tissue were used containing collagen gene, aggrecan and SOX9. The expression of gene confirmed by quality method of Reverse Transcriptase PCR then quantitatively examined by Real –Time PCR. Results: The finding showed that using nano fiber scaffold of poly lactic acid modified with glycosaminoglycan leads to the increase of expression of cartilage gene. Conclusion: Our findings indicated that GAG/PCL Nano fiber scaffold enhances the differentiation of mesenchymal stem cells into cartilage. This can help to overcome the limitations of tissue engineering.

Spontaneous Differentiation of Human Mesenchymal Stem Cells on Poly-Lactic-Co-Glycolic Acid Nano-Fiber Scaffold

INTRODUCTION Mesenchymal stem cells (MSCs) have immunosuppressive activity and can differentiate into bone and cartilage; and thus seem ideal for treatment of rheumatoid arthritis (RA). Here, we investigated the osteogenesis and chondrogenesis potentials of MSCs seeded onto nano-fiber scaffolds (NFs) in vitro and possible use for the repair of RA-affected joints. METHODS MSCs derived from hea...

Differentiation of Umbilical Cord Lining Membrane-Derived Mesenchymal Stem Cells into Endothelial-Like Cells

Background: Stem cell therapy for the treatment of vascular-related diseases through functional revascularization is one of the most important research areas in tissue engineering. The aim of this study was to investigate the in vitro differentiation of umbilical CL-MSC into endothelial lineage cells. Methods: In this study, isolated cells were characterized for expression of MSC-specific marke...

The effect of electrospun poly(lactic acid) and nanohydroxyapatite nanofibers’ diameter on proliferation and differentiation of mesenchymal stem cells

Objective(s): Electrospun nanofibrous mats of poly(lactic acid) (PLA) and nanohydroxyapatite (nano-HA) were prepared and proliferation and differentiation of mesenchymal stem cells on the prepared nanofibers were investigated in this study. Materials and Methods: PLA/nano-HA nanofibers were prepared by electrospinning. The effects of process parameters, such as nano-HA concentration, distance, ...

The differentiation of bone marrow mesenchymal stem cells into chondrocyte-like cells on poly-L-lactic acid (PLLA) scaffolds.

While intervertebral disc (IVD) degeneration is associated with the majority of cases of low back pain, current treatments are symptomatic rather than curative. Tissue engineering offers a treatment that both cures the problem of disc degeneration and restores normal disc function. One of the major problems for any tissue engineering strategy, however, is ensuring that both the cells and matric...

Differentiation of Mesenchymal Stem Cells Derived From Human Adipose Tissue into Cholinergic-like Cells: In Vitro Study

Introduction: Cholinergic-associated diseases currently constitute a significant cause of neurological and neurodegenerative disabilities.  As the drugs are not efficient in improving the suffered tissues, stem cell treatment is considered an effective strategy for substituting the lost cells. Methods: In the current study, we set out to investigate the differentiation properties of human adip...
