Abstract:

Background: The increasing use of nanoparticles (NPs) may have negative impacts on both organisms andthe environment. Objectives: The differential expression of mitochondrial manganese superoxide dismutase (MnSOD) gene in wheat in response to silver nitrate nanoparticles (AgNPs) and AgNO3 was investigated. Materials and Methods: A quantitative Real-Time RT-PCR experiment was carried out with MnSOD gene using RNAs isolated from wheat shoots treated for 0, 2, 6, 12, and 24 h with 100 mg.L-1 of either AgNO3 or AgNPs. Results: The results of this study showed that both treatments cause changes in the expression pattern of the MnSOD gene. While 2 and 6 h following the beginning of the stress, MnSOD expression was up-regulated significantly, in response to AgNO3 (1.4 and 2.8 fold, respectively), in response to AgNPs, it was up-regulated significant only after 6 h (1.6 fold), compared with the control. The gene expression, after 12 h in response to AgNO3 and AgNPs were down-regulated significantly (0.7 and 0.8 fold, respectively), and in the next 12 h , the expression appeared to be similar to the control. Conclusion: Exposure to both AgNPs and Ag ions led to a significant increase in MnSOD expression, but AgNO3 changed the MnSOD expression faster than AgNPs. Therefore, it is suggested that AgNO3 has greater penetrability and effectiveness.

Comparison of MAPK and thioredoxin gene expression in wheat seedlings exposed to silver nitrate and silver nanoparticle

The extensive use of heavy metals and nanoparticles (NPs) has led to their release into the environment that might have negative impacts on both organisms and the environment. In this study, the molecular responses of wheat seedlings to silver nitrate and silver nanoparticles (AgNPs) were assessed by transcript accumulation analysis of genes coding for products potentially involved in heavy met...

Morphological and Proteomic Responses of Eruca sativa Exposed to Silver Nanoparticles or Silver Nitrate

Silver nanoparticles (AgNPs) are widely used in commercial products, and there are growing concerns about their impact on the environment. Information about the molecular interaction of AgNPs with plants is lacking. To increase our understanding of the mechanisms involved in plant responses to AgNPs and to differentiate between particle specific and ionic silver effects we determined the morpho...

Isolation, chromosomal localization, and differential expression of mitochondrial manganese superoxide dismutase and chloroplastic copper/zinc superoxide dismutase genes in wheat.

Superoxide dismutase (SOD) gene expression was investigated to elucidate its role in drought and freezing tolerance in spring and winter wheat (Triticum aestivum). cDNAs encoding chloroplastic Cu/ZnSODs and mitochondrial MnSODs were isolated from wheat. MnSOD and Cu/ZnSOD genes were mapped to the long arms of the homologous group-2 and -7 chromosomes, respectively. Northern blots indicated that...

Catalase in Lung Cancer Differential Expression of Manganese Superoxide Dismutase

Reactive oxygen species (ROS) are important in the initiation and promotion of cells to neoplastic growth. In this context, cigarette smoke exposure, the primary risk factor in lung cancer development, leads to high levels of ROS within the human airway. Although well-equipped with an integrated antioxidant defense system consisting of low-molecular weight antioxidants such as glutathione and i...

Differential Phytotoxic Impact of Plant Mediated Silver Nanoparticles (AgNPs) and Silver Nitrate (AgNO3) on Brassica sp.

Continuous formation and utilization of nanoparticles (NPs) have resulted into significant discharge of nanosized particles into the environment. NPs find applications in numerous products and agriculture sector, and gaining importance in recent years. In the present study, silver nanoparticles (AgNPs) were biosynthesized from silver nitrate (AgNO3) by green synthesis approach using Aloe vera e...

Silver Nanoparticles from Ultrasonic Spray Pyrolysis of Aqueous Silver Nitrate

Silver particles less than 20 nm in diameter were synthesized by pyrolysis of an ultrasonically atomized spray of highly dilute aqueous silver nitrate solution at temperatures above 650◦C and below the melting point of silver. Feed solution concentration and ultrasound power applied to the atomizer were found to have a significant impact on the particle size of the silver nanoparticles. Average...
