Determination of malachite green and leucomalachite green residues in rainbow trout in Sanandaj City
Authors
Abstract:
Background and Aim: Malachite green (MG) is an organic compound which his used as a dye in textile industry. This compound is applied incorrectly and illegally in aquiculture as an antimicrobial agent. This compound has mutagenic and carcinogenic effects. Materials and Methods: In this study we determined the concentrations of MG and leucomalachite green (LMG) in the muscle tissue of 90 rainbow trouts (Oncorhynchus mykiss) collected from the local markets in Sanandaj City within a period of six months in 2018. In order to measure the residues of MG and LMG, we used enzyme-linked immunosorbent assay (ELISA) based on the European Union Decision 2002/657/EC. Results: The results showed presence of MG and LMG in all 90 samples: The residues of MG in the samples were as follows: 1-2 μg/Kg in 41 (6.45%), more than 2 μg/Kg in 45 (50%) and less than 1 μg/Kg in 4 samples (4.4%). Furthermore, the residues of LMG in the samples were as follows: more than 2 μg/Kg in 13 (45.14%), 1-2 μg/Kg in 67 (44.74%) and less than 1 μg/Kg in 10 samples (11.11%). Moreover, the limit of detection (LOD) in the method was 0.43 μg/Kg. Conclusions: It is concluded that MG is illegally applied in fish farms, which can be a threat to human health and the environment.

Reduction of malachite green to leucomalachite green by intestinal bacteria.
Intestinal microfloras from human, rat, mouse, and monkey fecal samples and 14 pure cultures of anaerobic bacteria representative of those found in the human gastrointestinal tract metabolized the triphenylmethane dye malachite green to leucomalachite green. The reduction of malachite green to the leuco derivative suggests that intestinal microflora could play an important role in the metabolic...
full textEnhanced Sensitive Immunoassay: Noncompetitive Phage Anti-Immune Complex Assay for the Determination of Malachite Green and Leucomalachite Green
To develop a more sensitive immunoassay for malachite green (MG) and leucomalachite green (LMG), we identified the immunocomplex binding phage-borne peptides for use in the noncompetitive phage anti-immunocomplex assay (PHAIA). An anti-LMG monoclonal antibody (mAb) was used to select immunocomplex binding peptides from a circular random eight-amino-acid phage-displayed library. After three roun...
full textEffects of Malachite Green and Its Major Metabolite, Leucomalachite Green, in Micromass Cultures of Rat Embryonic Cells
Embryonic mesenchyme limb (LB) and midbrain (CNS) cells cultured at high density were exposed to different concentrations of the fungicide malachite green (MG) and its major metabolite leucomalachite green (LMG). The concentration response curves were analysed, and two endpoints were calculated: the concentrations (μg/mL) that produced 50% inhibition of viability/proliferation (IC50-P) and diff...
full textEvaluation of the effects of different concentrations of neutral anolyte on fungal infected eggs in rainbow trout (Oncorhynchus mykiss) in comparison with green malachite
The objective of this study was to determine the effect of different concentrations of anolyte on saprolegniasis in comparison with green malachite in rainbow trout hatcheries, in the northern part of Iran, Tonekabon. Nearly 5000 green eggs of Oncorhynchus mykiss (equivalent to 300 g) were obtained from a private farm in the north of Iran. The study was designed in 5 treatments, 0.25, 0.5, 30 a...
full textDetermination of Malachite Green and Leucoma- Lachite Green in Trout and Carp Muscle by Liquid Chromatography with Visible and Fluorescence Detection
A fast and specific method for determination of malachite green (MG) and its major metabolite leucomalachite green (LMG) in trout and carp muscle is described. MG and LMG residues were extracted from fish muscle with an acetonitrile-buffer mixture and isolated by partitioning into dichloromethane. Extracts were then cleaned up on solid-phase extraction (SPE) columns. Chromatographic separation ...
