Design and Fabrication of a Novel Transplant Combined with Human Bone Marrow Mesenchymal Stem Cells and Platelet-rich Fibrin: New Horizons for Periodontal Tissue Regeneration after Dental Trauma
Authors
Abstract:
Avulsed teeth that are replanted dried are more prone to loss. Recent tissue engineering studies focus onfabrication of various cell delivery systems to improve the overall prognosis of such teeth. To evaluate this newcell transplant method, we initially aimed at designing of PRF scaffold and determining BMMSCs viabilityand function on the fabricated scaffold. To test this concept in-vitro, human BMMSCs were isolated andcharacterized by cell surface marker, and their potential on osteogenic/adipogenic differentiation. The biologicaleffects of PRF scaffold on human BMMSCs were then investigated by cell proliferation assay. The data werequantified for statistical analysis. It was found that PRF significantly induced BMMSCs proliferation throughoutthe incubation period due to its growth factor secretion. Results of MTT assay showed an increasing trend duringthe testing period from day 1 to day 7. The result of scanning electron microscopy showed that BMMSCs couldtightly adhere to fibrin scaffold just shortly after seeding. These data suggest that the BMMSCs/PRF constructhas the potential to improve the clinical prognosis of replanted avulsed teeth in future. Additional studies areneeded to be performed before its clinical use.

Study of platelet‐rich fibrin combined with rat periodontal ligament stem cells in periodontal tissue regeneration
The objective of this study was to investigate the advantages and feasibility of periodontal tissue regeneration using platelet-rich fibrin (PRF) combined with rat periodontal ligament stem cells (PDLSCs) for the first time. We first determined the effect of PRF on rat PDLSCs in vitro. We next conducted an in vivo study, in which a tissue engineering technique was performed to repair periodonta...

The use of platelet-rich fibrin combined with periodontal ligament and jaw bone mesenchymal stem cell sheets for periodontal tissue engineering
Periodontal regeneration involves the restoration of at least three unique tissues: cementum, periodontal ligament tissue (PDL) and alveolar bone tissue. Here, we first isolated human PDL stem cells (PDLSCs) and jaw bone mesenchymal stem cells (JBMSCs). These cells were then induced to form cell sheets using an ascorbic acid-rich approach, and the cell sheet properties, including morphology, th...
