Abstract:

The aim of the present study was to determine cytotoxic activity of crude methanolicextract of Echinophora platyloba on breast cancer MDA-MB-231 cell line. The free radicalscavenging effects of methanolic extract of E. platyloba were tested using DPPH method.Crude methanolic extract exhibited potential antioxidant activity with an IC50 value of 234.28 ±21.63 μg/mL when compared to the standard BHT with an IC50 value of the 19.5 ± 0.8 μg/mL.In addition, the in-vitro cytotoxic activity of this extract was studied against MDA-MB-231and MCF-10a cells by MTT assay for 12, 24 and 36 h. Our data showed 534.6 ± 7.2 μg/mL ofextract following 24 h of incubation was the most cytotoxic dose against MDA-MB-231 cellsin comparison with other doses. This extract could induce apoptosis and promote cell-cyclearrest at S-phase in MDA-MB-231 cells after 24 h of incubation, as compared to the controlgroup (p < 0.001) and could significantly up-regulate the expression of bax and p27 genes atthe level of 2.8 and 2.2 folds, respectively. While, a significant amount of down-regulation wasobserved for bcl-2 gene expression, which was observed to be 0.4 fold. The present resultsprove the anticancer capacity of crude methanolic extract of E. platyloba to inhibit limit cellproliferation, and inducing cell cycle arrest and apoptosis.

Quercetin induces cell cycle arrest and apoptosis in CD133+ cancer stem cells of human colorectal HT29 cancer cell line and enhances anticancer effects of doxorubicin

Objective(s):The colorectal cancer stem cells (CSCs) with the CD133+ phenotype are a rare fraction of cancer cells with the ability of self-renewal, unlimited proliferation and resistance to treatment. Quercetin has anticancer effects with the advantage of exhibiting low side effects. Therefore, we evaluated the anticancer effects of quercetin and doxorubicin (Dox) in HT29 cancer cells and its ...

Ganoderma lucidum extract induces cell cycle arrest and apoptosis in MCF-7 human breast cancer cell.

Although the pharmacology and clinical application of water extracts of Ganoderma lucidum have been extensively documented, little is known regarding its alcohol extract. In the present study, the anti-tumor effect of an alcohol extract of Ganoderma lucidum was investigated using MCF-7 cells. We found that the alcohol extract of Ganoderma lucidum inhibited cell proliferation in a dose- and time...

Acetonic Extract of Buxus sempervirens Induces Cell Cycle Arrest, Apoptosis and Autophagy in Breast Cancer Cells

Plants are an invaluable source of potential new anti-cancer drugs. Here, we investigated the cytotoxic activity of the acetonic extract of Buxus sempervirens on five breast cancer cell lines, MCF7, MCF10CA1a and T47D, three aggressive triple positive breast cancer cell lines, and BT-20 and MDA-MB-435, which are triple negative breast cancer cell lines. As a control, MCF10A, a spontaneously imm...

S-allylcysteine induces cell cycle arrest and apoptosis in androgen-independent human prostate cancer cells.

To increase the use of phytochemical supplements as chemoprevention or adjuvant drugs in cancer treatment, it is necessary to verify their biological effects and correlative mechanisms. Recently, S-allylcysteine (SAC) was identified as a potent compound derived from garlic. The aim of this study was to evaluate the anticancer effects of SAC on androgen-independent human prostate cancer (PC-3) c...
