Characterization of the Potential Probiotic Vibrio sp. V33 Antagonizing Vibrio Splendidus Based on Iron Competition
Authors
Abstract:
Background:Vibrio splendidusVs is an important aquaculture pathogen that can infect a broad host of marine organisms. In our previous study, an antagonistic bacterium Vibrio sp. V33 that possessed inhibitory effects on the growth and virulence of a pathogenic isolate V.splendidusVs was identified. Objectives: Here, we further explored the antagonistic substances and antagonistic effects from the viewpoint of iron competition. Materials and Methods: The main antagonistic substances in the supernatants from Vibrio sp. V33 were identified using the bioassay-guided method. The response of V. splendidus Vs under the challenge of cell-free supernatant from Vibrio sp. V33 was determined via sodium dodecyl sulfate-polyacrylamide gel electrophoresis and real-time reverse-transcription PCR. Results: The main antagonistic substances produced by Vibrio sp. V33 have low molecular weights, are water soluble, and are heat-stable substances. Meanwhile, the iron uptake rate of Vibrio sp. V33 was higher than that of V. splendidus Vs. In the presence of cell-free supernatant from Vibrio sp. V33, expressions of two functional genes, viuB and asbJ related to ferric uptake processes in V. splendidus Vs, were up-regulated, whereas furVs coding the ferric uptake repressor was suppressed below 0.5-fold. One gene coding phosphopyruvate hydratase does not change at mRNA level, but was up-regulated at protein level. Conclusions: Our results suggested that antagonistic effect of Vibrio sp. V33 on the pathogenic isolate V.splendidusVs was partially due to the stronger ability of Vibrio sp. V33 to seize iron. This cell-free supernatant from Vibrio sp. V33 created an iron-limited milieu for V. splendidusVs, which led to the changed expression profiles of genes that were related to iron uptake in V. splendidusVs.

Characterization of the alginate lyases from Vibrio 1 splendidus 12 B 01 2 3
18 Alginate lyases are enzymes that degrade alginate through β-elimination of the glycosidic bond 19 into smaller oligomers. We investigated the alginate lyases from Vibrio splendidus 12B01, a 20 marine bacterioplankton species that can grow on alginate as its sole carbon source. We 21 identified, purified, and characterized four polysaccharide lyase family 7 alginates lyases – 22 AlyA, AlyB, A...
full textDraft Genome Sequence of Vibrio splendidus DSM 19640
Here, we present the draft genome sequence of Vibrio splendidus type strain DSM 19640. V. splendidus is an abundant species among coastal vibrioplankton. The assembly resulted in a 5,729,362-bp draft genome with 5,032 protein-coding sequences, 6 rRNAs, and 117 tRNAs.
full textPhylogenetic study and identification of Vibrio splendidus-related strains based on gyrB gene sequences.
Different strains related to Vibrio splendidus have been associated with infection of aquatic animals. An epidemiological study of V. splendidus strains associated with Crassostrea gigas mortalities demonstrated genetic diversity within this group and suggested its polyphyletic nature. Recently 4 species, V. lentus, V. chagasii, V. pomeroyi and V. kanaloae, phenotypically related to V. splendid...
full textEffect of temperature, salinity and nutrient content on the survival responses of Vibrio splendidus biotype I.
The aim of this study was to evaluate the survival responses of two strains of Vibrio splendidus, both in natural and in defined media. For this purpose, freshwater and defined media containing different salinities (3.3-0.9 %) and nutrient concentrations (17-0.005 mg x l(-1)) were assayed. The incubation temperatures were established at 4, 10 and 22 degrees C. The acridine orange staining techn...
full textDraft Genome Sequence of the Oyster Larval Probiotic Bacterium Vibrio sp. Strain OY15
We report the draft genome sequence of Vibrio sp. strain OY15, a Gram-negative marine bacterium isolated from an oyster (Crassostrea virginica) digestive tract and shown to possess probiotic activity. The availability of this genome sequence will facilitate the study of the mechanisms of probiotic activity as well as virulence capacity.
full textIsolation of Vibrio alginolyticus and Vibrio splendidus from aquacultured carpet shell clam (Ruditapes decussatus) larvae associated with mass mortalities.
Two episodes of mortality of cultured carpet shell clams (Ruditapes decussatus) associated with bacterial infections were recorded during 2001 and 2002 in a commercial hatchery located in Spain. Vibrio alginolyticus was isolated as the primary organism from moribund clam larvae that were obtained during the two separate events. Vibrio splendidus biovar II, in addition to V. alginolyticus, was i...
