Changes of The Uterine Tissue in Rats With Polycystic Ovary Syndrome Induced by Estradiol Valerate

Objective : Polycystic ovary syndrome (PCOS) is one of the most common hormonal disorders that can lead to irregular menstrual cycles and hyperandrogenism. Reduced levels of progesterone and increased estrogen in these women can perpetually stimulate the endometrial tissue of the uterus. In this study, the effect of PCOS induction was studied using estradiol valerate. MaterialsAndMethods Adult female Wistar rats weighing approximately 200 gr were divided into control, sham, and experimental groups (n=6). In the experimental group, PCOS was induced using subcutaneous injection of 2 mg estradiol valerate. For histological and cytological studies, after the approval of syndrome induction, the rats in all groups were dissected and their uterus was removed and fixed. The uterine tissue sections were, then, stained with H&E and Iron- hematoxylin methods. Factors like height of the epithelium, thickness of the uterus wall, and frequency of the mitotic cells were examined. The data were assessed at α=0.05. Results Findings the study of uterine tissue revealed that the height of the uterus luminal epithelium, the thickness of the uterus wall and the frequency of eosinophils in the endometrial stroma were increased significantly in the experimental group. In addition, the frequency of mitotic cells in the experimental group was increased in both luminal and glandular epithelia of the uterus. The increasing rate of the glandular epithelium region was noticeable and significant. Conclusion The results indicate that induction of polycystic ovary syndrome by estrdiol valerate could change the proliferation rate in the endometrial tissue of the uterus.
