Calcitonin Gene-Related Peptide Effects on Phenotype and IL-12 Production of Monocyte-Derived Dendritic Cells in Rheumatoid Arthritis Patients
Authors
Abstract:
Objective(s) Recent studies on human indicate that the introduction of therapeutic use of tolerogenic dendritic cell (DC) for chronic inflammatory conditions is imminent. For the purpose of defining CGRP potency in tolerogenic DC production, we investigated the phenotype and IL-'2 production of DCs generated from the monocytes of rheumatoid arthritis (RA) patients in the presence of the calcitonin gene-related peptide (CGRP), as a multifunctional neuropeptide. Materials and Methods DCs were generated from isolated monocytes from four resistant and two early female RA patients using IL- 4, GM-CSF, and CGRP at concentrations of 0, ', and '00 nM. Then, the phenotype of neuropeptide-treated or untreated DCs was determined using flow cytometry and the IL-'2 production was measured by ELISA. Results Our study showed that, on the last day of the culture, at a concentration of ' nM CGRP, the mean fluorescence intensity (MFI) for CD80 increased ('4.'3%) and the MFIs for CD83, CD86, and HLA-DR decreased ('4.57%, 5.28%, and 6.88% respectively). Moreover, at '00 nM CGRP concentration, the MFI for CD80 increased (''.'0%) and the MFIs for CD83, CD86, and HLA-DR decreased (4.27%, '8.60%, and '9.75% respectively). In addition, our results indicated that the mean concentrations of IL-'2 produced at 0, ', and '00 nm CGRP concentrations measured '3.72±2.4', ''.0'±'.6', and 7±'.34 pg/ml respectively. Conclusion Decreased CD83, CD86, and HLA-DR expression and reduced IL-'2 production by CGRP were found in the RA patients' monocyte-derived DCs. CD83 is a well-defined DC activation marker. HLA-DR and CD86 are appropriate molecules for inducing an immune response. IL-'2 promotes cell-mediated immunity. Therefore we suggest that CGRP may be used as an inducer in the production of tolerogenic DCs.

full textDIFFERENTIATION OF MONOCYTE DERIVED DENDRITIC CELLS IN SERUM FREE CONDITIONS
Human peripheral blood monocytes (HPBM) were cultured in the absence of human serum and were converted into a state exhibiting a high accessory function expressed by their ability of supporting lymphocyte proliferation. After a prolonged culture in serum free media the monocyte derived cells were highly viable, increased in size and developed veils and dendritiform elongatio'l1s. Paralleli...
full textGeneration of Mature Monocyte-Derived Dendritic Cells in the Presence of Heparin and Monocyte Conditioned Medium: Phenotypic and Functional Comparison
Background: Dendritic cells (DC) induce tumor or pathogen-specific T cell responses in humans. Several laboratories have developed culture systems, including maturation factors for human DC from peripheral blood monocytes. We comprehensively compared standard maturation stimulus, an autologous monocyte-conditioned medium (MCM), with heparin for their ability to promote uniformly mature DC that ...
full textEvaluating the Effects of Cytomegalovirus Glycoprotein B on the Maturation and Function of Monocyte-derived dendritic cells
Background & Objectives: Interaction of cytomegalovirus glycoprotein B with toll-like receptors of dendritic cells leads to early signaling and innate immune responses. The aim of this study is to evaluate the effects of cytomegalovirus glycoprotein B on the maturation and function of monocyte-derived dendritic cells in treated groups in comparison with control groups. Materials & Methods: Bloo...
full textMaturation State and Function of Monocyte Derived Dendritic Cells in Liver Transplant Recipients
Background: Dendritic cells (DCs) are potent antigen presenting cells for triggering of the immune reaction post transplantation. These cells are centrally involved in the initiation of T cell-dependent immune responses. Objective: To compare the level of DC maturation and function in liver transplant recipients with healthy controls. Methods: In this study, twelve peripheral blood samples we...
full textIL-12 responsiveness and expression of IL-12 receptor in human peripheral blood monocyte-derived dendritic cells.
We analyzed the expression of IL-12Rbeta1 and IL-12Rbeta2 and the role of IL-12 in the activation of monocyte-derived dendritic cells (DCs) via IL-12Rbeta1-mediated signaling events. Flow cytometric analysis revealed that IL-12Rbeta1 was expressed in T cells, Con A blasts, and monocyte-derived DCs, but not in monocytes, while its transcript was detected in all of these cell types. Transcription...
