Abstract:

Catharanthus roseus is a medicinal and ornamental plant with growing attention toward its economical value. Cell suspension from C. roseus were treated with 0 to 60mM of cadmium nitrate for 1, 3 and 6 days, then cell viability was determined using trypan blue and MTT assay. Cell morphology was investigated using 0, 10, 30 and 50 mM of cadmium nitrate as selected dose for 3 days. Callus of C.roseus was treated with selected doses for same period to estimate the hydrogen peroxide content, level of lipid peroxidation, proline content and the activity of superoxide dismutase, catalase and peroxidase as well as protein profile. In addition, the production of total alkaloid, flavonoid and phenolic compounds was estimated. Data was analyzed using one-way ANOVA, Duncan test and p

Improved alkaloid content in callus culture of Catharanthus roseus

The low yield and high market price of the pharmaceutically important alkaloids of Catharanthus roseus (L.) G. Don viz. vincristine, vinblastine and ajmalicine have created interest in improved alternative routes for their production such as using cell and tissue culture. The callus developed on Murashige and Skoog (MS) media supplemented with different concentrations of auxins and cytokinins w...

5-Methyltryptophan resistant cells of Catharanthus roseus.

Several cell lines resistant to 5-methyltryptophan were selected from wild type cells of different Catharanthus roseus suspension cultures. The resistant cells had up to 30 times the normal levels of free tryptophan. Despite the increased pool size of tryptophan anthranilate synthetase activity of resistant cells was as sensitive to inhibition by L-tryptophan as wild type cells. The overproduct...

Exogenous Nitric oxide (NO) induced oxidative stress and increased production of secondary metabolites in Catharanthus roseus Callus Cells

The objective of the study was to investigate the effect of exogenous nitric oxide on cellular response and production of secondary metabolites in Catharanthus roseus callus. The Cell suspension and callus of C. roseus were treated with sodium nitroprusside, then cell viability, morphology, the amount of H2O2, proline, lipid peroxidation, the activity of superoxide dismutase, catalase and perox...

Optimization of callus induction and cell suspension cultures of Catharanthus roseus

Background & Aim: Investigation of suspension cell culture and callus production in Catharanthus roseus. Catharanthus roseus is an herbaceous perennial plant belongs to the family Apocynaceae. C. roseus is important as commercial medicinal plant because of its alkaloidal content. Whole plant contains about 130 alkaloids those are classified as vinca alkaloids...
