AXENIC CULTURE AND CRYOPRESERVATION OF GIARDIA LAMBLIA ISOLATED IN IRAN
Authors
Abstract:
Giardia lamblia cysts were isolated from fecal samples of 55 symptomatic and asymptomatic patients with giardiasis. The cysts were harvested and purified using the sucrose gradient method. IS The excystation of G. lamblia was obtained by treating them with HCI (0.01N)', then they were transferred on to modified TYI-S-33 medium. Out of 55 stool samples 17 cases were excysted with a rate of 4-32%,but we were able to cultivate only one case with 12% excystation in culture medium. The generation time of G.lamblia was calculated to be about 10.33 hours. Cryopreservation of trophozoites was successfully done in liquid nitrogen and the thawing process had a survival rate of 85-90%.

Introduction: Giardia lamblia is a protozoan parasite with universal distribution in human populations. This infections transfer to human via contaminated foods and waters with Giardia cysts. The Knowledge on the incidence of this agent in the potential infection sources can provide valuable information for control and the spread of this parasite to human communities. This study was aimed to is...
full textGiardia lamblia and Giardiasis
Introduction Ask the average outdoors person about Giardia lamblia, and they have certainly heard about it. Almost always, however, they are considerably misinformed about both the organism’s significance in wilderness water and the seriousness of the disease, giardiasis, if contracted. The amount of information easily found on the subject is voluminous. Unfortunately, almost all of it is flawe...
full textBiology of Giardia lamblia.
Giardia lamblia is a common cause of diarrhea in humans and other mammals throughout the world. It can be distinguished from other Giardia species by light or electron microscopy. The two major genotypes of G. lamblia that infect humans are so different genetically and biologically that they may warrant separate species or subspecies designations. Trophozoites have nuclei and a well-developed c...
full textPyrimidine salvage in Giardia lamblia
We have found that the anaerobic protozoan parasite Giardia lamblia is incapable of de novo pyrimidine metabolism, as shown by its inability to incorporate orotate, bicarbonate, and aspartate into the pyrimidine nucleotide pool. Results from high performance liquid chromatography of pyrimidine and pyrimidine nucleoside pulse-labeled nucleotide pools and enzyme assays suggest that the parasite s...
