Abstract:

Abstract : Foot and Mouth Disease is one of  the important live stocks contagious viral disease caused by Aphtovirus genus ,  that is belong to family RNA virus  picornaviride. The important characteristic of FMD virus is high mutation that give rise to diversity of Antigen in surface of Neutralizing proteins. For this reason FMD virus have 7 distinct serotype and many subtype. Vaccination is one of the methods for control disease in order to different type and subtype  of FMD virus , the effective vaccine should have virus that closed relationship with field virus and also many passage on cell culture may cause mutation on viral genome . The rate of genetic changes of FMD virus type O2016 during of 70 passages in BHK monolayer  cell is the main reasone of this paper .  After determine the type of virus in epithelium sample by ELISA , the virus inoculated in BHK monolayer cell for 70 consequence passage . harvested viruses in 10 selected passage (P1,P10 , P20,P30,P40,P50,P60,P70) is done by RT-PCR , ELISA , titration , real time PCR, Double dimension virus neutralization test for  immunological relationship value (r value) and Nucleotide sequencing of 1D segment  of viral genome . harvested FMD virus type O2016  From passage 1 until 70  be constant and no significant change neither in Sequencing nor in r value .

Rapid selection in modified BHK-21 cells of a foot-and-mouth disease virus variant showing alterations in cell tropism.

With persistent foot-and-mouth disease virus (FMDV) in BHK-21 cells, there is coevolution of the cells and the resident virus; the virulence of the virus for the parental BHK-21 cells is gradually increased, and the cells become partially resistant to FMDV. Here we report that variants of FMDV C3Arg/85 were selected in a single infection of partially resistant BHK-21 cells (termed BHK-Rb cells)...

Genetic stability of foot-and-mouth disease virus during long-term infections in natural hosts

Foot-and-mouth disease (FMD) is a severe infection caused by a picornavirus that affects livestock and wildlife. Persistence in ruminants is a well-documented feature of Foot-and-mouth disease virus (FMDV) pathogenesis and a major concern for disease control. Persistently infected animals harbor virus for extended periods, providing a unique opportunity to study within-host virus evolution. Thi...

Organic Circulation and persistence of Foot – and – Mouth disease virus type O in guinea pig

Since, the most susceptible laboratory animal to Foot–and–Mouth disease virus (FMDV) is guinea pig, and then one milliliter of FMDV type (O) concentrations of 106-106.5 TCID50was inoculated intradermally (ID) to 10 plantar surface of guinea pig (right side). Guinea pig adapted virus has been prepared after generalization phase, and then one milliliter of virus was inoculated intradermally to 30...

Nucleotide mismatches of foot-and-mouth disease virus during replication.

As there is a lack of error correction mechanisms during RNA replication, foot-and-mouth disease virus (FMDV) has a very high mismatch rate, which leads to a high mutation rate, in the range of 10(-3) to 10(-5) per nucleotide site per genome replication. We examined the nucleotide mismatch of FMDV during replication, based on the whole genomes of the 7 serotypes retrieved from NCBI. With the Me...
