Apoptosis induction of human endometriotic epithelial and stromal cells by noscapine
Authors
Abstract:
Objective(s): Endometriosis is a complex gynecologic disease with unknown etiology. Noscapine has been introduced as a cancer cell suppressor. Endometriosis was considered as a cancer like disorder, The aim of present study was to investigate noscapine apoptotic effect on human endometriotic epithelial and stromal cells in vitro. Materials and Methods:In this in vitro study, endometrial biopsies from endometriosis patients (n=9) were prepared and digested by an enzymatic method (collagenase I, 2 mg/ml). Stromal and epithelial cells were separated by sequential filtration through a cell strainer and ficoll layering. The cells of each sample were divided into five groups: control (0), 10, 25, 50 and 100 micromole/liter (µM) concentration of noscapine and were cultured for three different periods of times; 24, 48 and 72 hr. Cell viability was assessed by colorimetric assay. Nitric oxide (NO) concentration was measured by Griess reagent. Cell death was analyzed by Acridine Orange (AO)–Ethidium Bromide (EB) double staining and Terminal deoxynucleotidyl transferase (TdT) dUTP Nick-End Labeling (TUNEL) assay. Data were analyzed by one-way ANOVA. Results: Viability of endometrial epithelial and stromal cells significantly decreased in 10, 25, 50 and 100 µM noscapine concentration in 24, 48, 72 hr (P

Background: Endometriosis is the presence of a functional endometrial tissue outside the uterine cavity and defines as cancer like model. Noscapine is a safe cough suppressant which has been introduced as cancer suppressor. The aim of this study was to investigate the in vitro effect of noscapine on human endometriotic epithelial cells. Materials and Methods: Human endometriotic biopsies (n=7) ...
full textP-99: Survey of Noscapine Effect on Apoptosis of Human Endometrial Stromal Cells: An In Vitro Study
Background: Noscapine is a water soluble alkaloid with antitussive property and very less toxicity. It is obtained from the Papaveraceae family. Recently, new pharmacological properties such as anticancer, anti stroke and anti-angiogenic effects are introduced. There is no scientific report on noscapine effect on reproductive system. The aim of present study was to determine in vitro effect of ...
full textP-136: High Concentrations of Noscapine Induces Apoptosis in Stromal Cells of Endometriosis In Vitro
Background: Endometriosis is a benign hormone-dependent disease in women of reproductive age. There is no cure for eradicating the disease and there is need for new drugs. The aim of this study was to investigate the in vitro effect of noscapine as inducer of cell death on endometriotic stromal cells in patient's endometriosis in vitro. Materials and Methods: Endometrial biopsies from pati...

BACKGROUND The survival of endometriotic cells in the ectopic site has been investigated from the aspect of susceptibility of endometriotic tissues to apoptosis. In order to investigate the nature of abnormal survival of endometriotic cells in ectopic locations, we compared drug-induced apoptosis in endometrial and endometriotic cells. METHODS Endometrial stromal cells were obtained from norm...
