Anticonvulsant effects of intrahippocacampal N6-cyclohexyladenosine on piriform cortex-kindled seizures
Authors
Abstract:
In this study the role of adenosine A1 receptors of the CA1 region of hippocampus on piriform cortex–kindled seizures was investigated in rats. Obtained results showed that in kindled animals, bilateral microinjection of N6–cyclohexyladenosine (CHA), an adenosine A1 receptor agonist, at doses of 10 and 100 mM into the hippocampal CA1 region decreased the afterdischarge duration (ADD), stage 5 seizure duration (S5D) and seizure duration (SD), and increased the latency to stage 4 (S4L) significantly. CHA at the doses of 1 mM decreased only ADD and no changes were observed at the doses of 0.1 mM. Bilateral microinjection of 1,3-dimethyl-8-cyclopenthylxanthine (CPT), an adenosine A1 receptor antagonist, into the CA1 region of hippocampus increased ADD at the dose of 20 mM significantly, but has no effect on seizure parameters at the dose of 10 mM. Pretreatment of animals with CPT (10 mM) before CHA (10 and 100 mM), reduced the effect of CHA on seizure parameters significantly. Thus, it may be suggested that the hippocampal CA1 region may play a role in relaying or spreading of seizure spikes elicited from the piriform cortex, and activation of adenosine A1 receptors of this region can prevent this actions.

The effect of parameters of low-frequency electrical stimulation on piriform-cortex kindled seizures in rat
Introduction: Electrical low-frequency stimulation (LFS) has antiepileptic effect, but the role of different stimulation parameters on this effect has not been determined. In this study the effect of different LFS parameters (intensity, pulse duration and train duration) on piriform-cortex kindled seizures was investigated. Methods: Seizure was produced in animals using kindling model of ep...

Background & Aim: Hippocampal CA1 region is the most common area in the development of temporal lobe epilepsy and piriform cortex is recognized as an important structure involved in the control of kindled seizures. This fundamental study dealt with the role of adenosine A1 receptors of piriform cortex in CA1 hippocampal kindled seizures in rats. Material & Method: We implanted a tripolar el...

The effects of intraperitoneal injection of N6-cyclohexyladenosine (CHA, a selective adenosine A1 receptor agonist) and 8-cyclopenthyle-I-3-dimethylexanthine (CPT, a selective adenosine A1 receptor antagonist) on entorhinal cortex-kindled seizures were investigated. Fully entorhinal cortex-kindled rats received normal saline (control), CHA (0.06, 0.12 and 0.25 mg/kg) or CPT (0.06 and 0.12 mg/kg...

INTRODUCTION The hippocampus and piriform cortex have a critical role in seizure propagation. In this study, the role of adenosine A1 receptors of piriform cortex on CA1 hippocampal kindled seizures was studied in rats. METHODS Animals were implanted with a tripolar electrode in the right hippocampal CA1 region and two guide cannulae in the left and right piriform cortex. They were kindled by...
