A Preliminary Investigation of the Jack-bean Urease Inhibition by Randomly Selected Traditional used Herbal Medicine

Abstract:
Helicobacter pylori :28 µg/mL). (H. pylori) infection leads to different clinical and pathological outcomes in humans, including chronic gastritis, peptic ulcer disease and gastric neoplasia and even gastric cancer and its eradiation dependst upon multi-drug therapy. The most effective therapy is still unknown and prompts people to make great efforts to find better and more modern natural or synthetic anti-H. pylori agents. In this report 21 randomly selected herbal methanolic extracts were evaluated for their effect on inhibition of Jack-bean urease using the indophenol method as described by Weatherburn. The inhibition potency was measured by UV spectroscopy technique at 630 nm which attributes to released ammonium. Among these extracts, five showed potent inhibitory activities with IC50 ranges of 18-35 µg/mL. These plants are Matricaria disciforme (IC50:35 µg/mL), Nasturtium officinale (IC50:18 µg/mL), Punica granatum (IC50:30 µg/mL), Camelia sinensis (IC50:35 µg/mL), Citrus aurantifolia (IC50

full textKinetics and mechanism of jack bean urease inhibition by Hg2+
UNLABELLED BACKGROUND Jack bean urease (EC 3.5.1.5) is a metalloenzyme, which catalyzes the hydrolysis of urea to produce ammonia and carbon dioxide. The heavy metal ions are common inhibitors to control the rate of the enzymatic urea hydrolysis, which take the Hg2+ as the representative. Hg2+ affects the enzyme activity causing loss of the biological function of the enzyme, which threatens ...
full textKinetics and Mechanism Study of Competitive Inhibition of Jack-Bean Urease by Baicalin
Baicalin (BA) is the principal component of Radix Scutellariae responsible for its pharmacological activity. In this study, kinetics and mechanism of inhibition by BA against jack-bean urease were investigated for its therapeutic potential. It was revealed that the IC₅₀ of BA against jack-bean urease was 2.74 ± 0.51 mM, which was proved to be a competitive and concentration-dependent inhibition...

Investigation of their ultracentrifugal and electrophoretic behavior and chemical reactivity has been used to characterize the suprastructure of the nongenetic urease isozymes. As purified from jack bean meal, urease is obtained in the LY form (M, = 480,000, s20,W = 18.3), which is composed of two noncovalently bound, equal and active A1 half-units (M, = 240,000, sZo,W = 11.5) which separate af...
