روش کشت تخمدان بالغ در تکگثیر جنسی گیاه Argania spinosa L.SKeels

Abstract:

روش درون شیشه ای کشت تخمدان به منظور تولید گیاه Argania spinosa L.   sKeels  و افزایش درصد جوانه زنی بذور آن به کار گرفته شد. ضدعفونی سطحی بذور با اتانول 70% شستشو با آب مقطر استریل انجام شد. تخمدانها پس از حذف درون بر (اندوکارپ) سخت میوه ایزوله شده است. تخمدانها پس از ضدعفونی سطحی در قالب دو تیمار، کامل و برش دارشده در محیطهای کشت جامد wpm،ws  وHalf-mکشت شدند. بیشترین درصد جوانه زنی جنین در قالب کشت تخمدانهای برش دار شده در همه محیطهای کشت مورد استفاده اتفاق افتاد(63/62%) علیرغم اینکه بالاترین درصد جوانه زنی جنین در محیط کشت msمشاهده شده است52/49%) لیکن اختلاف معنی داری بین محیط های مختلف کشت در درصد جوانه زنی جنین درهر دو شیوه درهردو شیوه کشت تخمدانهای سالم وبرش دار شده دیده نشده است. کیاهان حاصل پس ازانجام موفق سازگاری تدریجی به گلخانه انتقال یافتند.
