(Daucus carota L.)
Authors
Abstract:
Experiments were carried out for optimization of conditions for morphogenic callus production and regeneration of carrot (Daucus carota L.) using four cultivated cultivars in Iran. Hypocotyl segments were cultured on medium supplemented with 0.2 mg/l 2,4-D for callus initiation and proliferation. Small portions of calli (25 mg) were cut and transferred to new media with different concentrations of 2,4-D including 0.2, 0.5 and 1 mg/l. Results showed that low level of 2,4-D was more effective in morphogenic callus production although its high concentrations caused more callus growth. Kinetin in concentration of 0.1 mg/l was also more effective for regeneration than benzyl amino purine (1 mg/l), especially according to embryoid production. MS and MSm media were used in the experiments to identify the differences between these two media according to callus, embryoid and plantlet production. Obtained results showed that MSm is more useful than MS medium for callus production and regeneration of carrot plantlets. Among tested cultivars improved Nantes had more capability to produce viable plantlets.

full textPervasive Mitochondrial Sequence Heteroplasmy in Natural Populations of Wild Carrot, Daucus carota spp. carota L
Exceptions to the generally accepted rules that plant mitochondrial genomes are strictly maternally inherited and that within-individual sequence diversity in those genomes, i.e., heteroplasmy, should be minimal are becoming increasingly apparent especially with regard to sequence-level heteroplasmy. These findings raise questions about the potential significance of such heteroplasmy for plant ...

full textCharacterization of Inositol Phosphates in Carrot (Daucus carota L.) Cells.
We have shown previously that inositol-1,4,5-trisphosphate (IP(3)) stimulates an efflux of (45)Ca(2+) from fusogenic carrot protoplasts (M Rincón, WF Boss [1987] Plant Physiol 83: 395-398). In light of these results, we suggested that IP(3) might serve as a second messenger for the mobilization of intracellular Ca(2+) in higher plant cells. To determine whether or not IP(3) and other inositol p...
full textGuaiacol Peroxidases in Carrot (Daucus carota L.) Root**
Carrot (Daucus carota L.) belongs to the group of common edible vegetables. The aim of the present study was to establish the distribution of guaiacol peroxidases in carrot root sections as well as to determine the enzymatic activity and the electrophoretic pattern of isoperoxidases in tissue extracts of particular root regions: central cylinder, inner cortex and epidermis, including the periph...
full textInositol Trisphosphate Metabolism in Carrot (Daucus carota L.) Cells.
The metabolism of exogenously added d-myo-[1-(3)H]inositol 1,4,5-trisphosphate (IP(3)) has been examined in microsomal membrane and soluble fractions of carrot (Daucus carota L.) cells grown in suspension culture. When [(3)H]IP(3) was added to a microsomal membrane fraction, [(3)H]IP(2) was the primary metabolite consisting of approximately 83% of the total recovered [(3)H] by paper electrophor...
