Abstract:

Background: Pancreatic islet transplantation has been reported as an appropriate method for treatment of type I diabetes patients, however there are strong indications that cytokine and chemokines secreted from transplanted islets play an important role in islet graft rejection in different stage post-transplantation. The NF-kB signaling pathway is activated in response to the stress resulted from isolation and purification process of pancreatic islets. Secretion and release of inflammatory mediators, including MCP-1, result from activation of this pathway which plays important part in activation of inflammatory processes accelerating graft rejection. Methods: This study was performed to examine the effect of curcumin on secretion of inflammatory mediators and function of pancreatic islets. Results: We observed that curcumin significantly decreased MCP-1 release from mouse islets compared to the control group and had no effect on function of pancreatic islets. Conclusion: Investigating the stimulatory signals leading to production and secretion of inflammatory mediators from pancreatic islets and discovering their underlying mechanisms will be useful in finding new therapeutic interventions for blocking inflammatory pathways and improvement in outcome of islet cell transplantation.

NF-κB signaling.

The NF-κB transcription factor is a critical regulator of the immune system, and is responsive to a large number of stimuli. Different stimuli engage signaling pathways to activate NF-κB, and effect distinct cellular responses. Mathematical modeling of the NF-κB network has been useful in studying the dynamic and cross-talk regulation of NF-κB. In this review, we discuss the regulation of NF-κB...

Hypoxia Induced NF-κB.

As Nuclear Factor-κB (NF-κB) is a major transcription factor responding to cellular stress, it is perhaps not surprising that is activated by hypoxia, or decreased oxygen availability. However, how NF-κB becomes activated in hypoxia is still not completely understood. Several mechanisms have been proposed and this review will focus on the main findings highlighting the molecules that have been ...

NF-κB in premature aging

consequence of a stochastic process caused by the accumulative effect of damaged molecules. However, recent experimental evidences have extended this view and suggested that aging also requires active signaling programs for the maintenance of the aged state [1]. Beyond cell-autonomous alterations, age signals get systemic through changes in intercellular communication pathways [2]. The identifi...
