ارزیابی بیان فیمبریه (Fim2 و Fim3) Bordetella pertussis در مراحل مختلف کشت به منظور تولید واکسن

Abstract:
زمینه و هدف: فیمبریه(Fim2 و Fim3) از فاکتورهای حدت در Bordetella pertussis است و باعث اتصال باکتری به سلولهای مژه دار دستگاه تنفس میزبان و تحریک پاسخهای ایمنی میشود. فیمبریه از ارکان اصلی واکسنهای سلولی و بدون سلولی سیاه سرفه محسوب میشود. طبق پیشنهاد سازمان بهداشت جهانی (WHO) در واکسنهای سلولی باید حضور فیمبریه (Fim2 و Fim3) تایید شده باشد. در این تحقیق برای بررسی و تایید حضور Fim2 و Fim3 در واکسنهای سلولی، دو سویهی واکسینال 509 B. pertussis(دارای Fim2) و 134 B. pertussis (دارای Fim3) مورد آزمایش قرار گرفتند. سپس میزان بیان Fim2 و Fim3 در مراحل مختلف کشت برای تولید واکسن در بیوراکتور مورد ارزیابی قرار گرفتند. روش بررسی: برای تعیین و ارزیابی حضور فیمبریه ها از روش اسلاید آگلوتینوژن و ELISA غیر مستقیم استفاده شد و مقدار آن به صورت کمی محاسبه گردید. پس از تایید حضور فیمبریه ها دو سویه در محیط مایع B2 کشت داده شدند. یافتهها: میزان بیان فیمبریه ها در فاز های مختلف رشد به دست آمد و بهترین زمان برای برداشت و توقف کشت تعیین گردید. نتیجهگیری: طبق نتایج بیشترین میزان بیان Fim2 و Fim3 و بهترین زمان برای برداشت باکتری از فرمانتور فاز لگاریتمی باکتری است. با اینکه میزان بیان این دو فیمبریه در این فاز با یکدیگر متفاوت می باشد اما در نهایت بیشترین حضور فیمبریه در این فاز است.

The fim genes which code for the fimbria protective antigens present in both the inactivated whole-cell and acellular vaccines were analyzed in 86 Canadian Bordetella pertussis isolates.
