abstract

genome-wide association studies (gwas) have identified genetic variants contributing to the risk of cardiovascular disease (cvd) at the chromosome 9p21 locus. the chromosome 9p21 is an important susceptibility locus for several multifactorial diseases like ischemic stroke, aortic aneurysm, type 2 diabetes mellitus and coronary artery disease (cad). f7 gene because of its role in activating the extrinsic pathway by the exposure of tissue factor after plaque disruption is related to atherothrombosis. the aim of the present study was to evaluate methylation status of two cad related genes, p15ink4b and f7, in iranian patients with coronary artery disease (cad). thirty samples from 15 male and 15 female with diagnosed 3 vessels disease cad and 60 samples from 60 non-cad controls who underwent coronary angiography was analyzed by msp and direct sequencing. dna methylation levels at p15ink4b gene increased significantly in cad patients in comparison with control group (p-value

Standard DNA methylation analysis in mouse epidermis: bisulfite sequencing, methylation-specific PCR, and 5-methyl-cytosine (5mC) immunological detection.

In mammals, methylation of cytosine C-5 position is a major heritable epigenetic mark on the DNA molecule. Maintenance of proper DNA methylation patterns is a key process during embryo development and in the maintenance of adult tissue homeostasis. The use of experimental procedures based on the chemical modification of cytosine by sodium bisulfite and the development of antibodies recognizing ...

Two Steps Methylation Specific PCR for Assessment of APC Promoter Methylation in Gastric Adenocarcinoma

Gastric Cancer (GC) is the second most common cancer in the world and a leading cause of cancer-related mortality. Methylation of promoter CpG islands (CGIs) belonging to tumor suppressor genes causes transcriptional silencing of their corresponding genes leading to carcinogenesis and other disorders. Adenomatous Polyposis Coli (APC) a tumor suppressor gene is inactivated by methylation of prom...

Methylation of p15INK4b and Expression of ANRIL on Chromosome 9p21 Are Associated with Coronary Artery Disease

BACKGROUND Genome-wide association studies have identified that multiple single nucleiotide polymorphisms on chromosome 9p21 are tightly associated with coronary artery disease (CAD). However, the mechanism linking this risk locus to CAD remains unclear. METHODOLOGY/PRINCIPAL FINDINGS The methylation status of six candidate genes (BAX, BCL-2, TIMP3, p14(ARF), p15(INK4b) and p16(INK4a)) in 205...

Single-cell, locus-specific bisulfite sequencing (SLBS) for direct detection of epimutations in DNA methylation patterns

Stochastic epigenetic changes drive biological processes, such as development, aging and disease. Yet, epigenetic information is typically collected from millions of cells, thereby precluding a more precise understanding of cell-to-cell variability and the pathogenic history of epimutations. Here we present a novel procedure for directly detecting epimutations in DNA methylation patterns using ...
