antispasmodic and antidiarrhoeal activities of 6-(4-hydroxy-3-methoxyphenyl)-hexanonic acid from pycnocycla spinosa decne. exboiss

abstract

pharmacological activities of 6-(4-hydroxy-3-methoxyphenyl)-hexanonic acid (hmpha), a phenolic compound, isolated from the extract of pycnocycla spinosa was investigated on ileum motility in vivo and in vitro . ileum motility was examined by measuring charcoal movement through the gut in mice. in addition, antidiarrhoeal activity of hmpha was assessed and compared with standard drug; loperamide (2 mg/kg) and the hydroalcoholic extract of p. spinosa (2 mg/kg). furthermore, concentration response curve to contraction induced by acetylcholine (ach), 5-hydroxy triptamine (5-ht) and electrical field stimulation (efs) were obtained after incubation of ileum segment with various concentrations of hmpha or hydroalcoholic extract of p. spinosa . hmpha (2 mg/kg and 5 mg/kg, orally) significantly inhibited gut movements in vivo and reduced diarrhoea induced by castor oil or sulphate magnesium. in addition, hmpha reduced ileum contraction induced by ach (ic 50 =33 ± 6 µg/ml), 5-ht (ic 50 =87 ± 12 µg/ml) and efs (ic 50 =36 ± 3 µg/ml) in vitro in a concentration-dependent manner. the inhibitory effect was reversible following washing off the drug. these studies indicate that hmpha as an active component of p. spinosa extract has significant antispasmodic and antidiarrhoeal activities and therefore, has the potential as a lead compound for further development of a new spasmolytic remedy.

Diurnal Variation of Essential of the Oil Components of Pycnocycla spinosa Decne. ex Boiss

BACKGROUND Pycnocycla spinosa Decne. ex Boiss is an aromatic plant which showed relaxant effects on isolated ileum contractions and antidiarrheal activity. Thirty four components have been extracted from P. spinosa essential oil, of which several major constituents were found to show seasonal variation. OBJECTIVES The aim of this work is to evaluate the diurnal variation of its oil constituen...

SYNTHESIS OF 8-HYDROXY-6-METHOXY- 3-UNDECYLISOCOUMARIN AND 2-HY DROXY - 4-METHOXY-6-(2-OXOTRIDECYL) BENZOIC ACID

Straightforward conversion of (+)-6,8-dimethoxy-3,4-dihydro-3- undecylisocoumarin (3) to the title isocoumarin was carried out. Hydrolytic ring opening of (3) afforded the hydroxy acid (4) which was immediately oxidized to keto acid (5) using chromic acid, Cyclodehydration of (5) afforded the 6,8- dimethoxy-3-undecylisocoumarin (6) which on selective demethylation of 8- methoxy group furni...

Seasonal Variation of Mono- and Sesquiterpenes in the Essential Oil of Pycnocycla Spinosa Decne. ex Boiss.

Pycnocycla spinosa aerial parts were collected weekly during four-month growth. Hydrodistillation and gas chromatography coupled with mass spectroscopy techniques were used for essential oil and sesquiterpenes investigation. Thirty-four components were identified, of which the characteristic sesquiterpenes were a -copaene, caryophyllene, a -humulene, b-ionene, d-cadinene, a -calacorene, caryoph...
