abstract
background: leishmaniasis is a group of diseases that are created by intracellular parasites of leishmania. cutaneous leishmaniasis is considered as one of the health problems in some provinces of iran. methods: in this study, a total of 178 giemsa-stained slides from confirmed cases of cutaneous leishmaniasis were examined. the slides were prepared from the patients with cutaneous leishmaniasis that referred to health centers and infected during the epidemic of cutaneous leishmaniasis in poldokhtar city, lorestan province, iran in 2006.genomic dna from each slide was extracted. after dna extraction, its-pcr was used. results: out of 178 slides, 129 (72.47%) samples had a band in the range of 485 bp and 49 (27.53%) samples 626 bp that matched l. tropica and l. major standard samples, respectively. conclusion: this study showed that leishmania dna could be efficiently ex-tracted and amplified even from old giemsa-stained microscopic slides that were stored more than 6 yr. in this study was shown that both l. tropica and l. major species exist in lorestan province.

full textDNA extraction and amplification of leishmania from archived, Giemsa-stained slides, for the diagnosis of cutaneous Leishmaniasis by PCR.
DNA was isolated from 92 Giemsa-stained smears of lesions from suspected cases of cutaneous leishmaniasis and used for PCR-based diagnosis of Leishmania infection. Each smear had been examined under a light microscope at x 1,000 and scored for amastigote numbers. Although the smears had been stored for up to 4 years, all the microscopy-positive slides were also positive by PCR and four of the 1...

Background: Direct identification of Leishmania species in Giemsa-stained slides without parasite culturing in the areas where multiple species exist, is very helpful. This study was designed to isolate Leishmani spp. from Giemsa-stained smears and to characterize them by PCR technique. Methods: A total of 48 Giemsa-Stained slides from confirmed cases of leishmaniasis were examined under a ligh...

full textIdentification of Leishmania species isolated from paitients with cutaneous leishmaniasis in Ghohab Mohammad Abad Isfahan using PCR method
Background and aim: Cutaneous leishmaniasis (CL) is a health problem of many countries in tropical and subtropical regions including Iran. Isfahan province is one of the foci of CL with the highest prevalence in Iran. This study was done to identify the species of Leishmania isolated from the patients in Ghohab Mohammad Abad, located in southwest of Isfahan which is a new foci of CL. Identifica...

BACKGROUND To determine the epidemiological status of cutaneous leishmaniasis outbreak, isolation and identification of the agent parasite, Leishmania, using PCR method in Gonbad-e Qabus County, north Iran, during 2006-2007. METHODS Data were collected on the prevalence of scars and ulcers over a period of 3 months among 6990 inhabitants of five villages around Gonbad-e Qabus County, north Ir...
