the effects of nitric oxide on gentamicin toxicity in isolated perfused rat kidney
Authors
abstract
introduction: there have been many studies in recent years on the role of nitric oxide (no) in acute renal failure. in this study, the effects of inhibition or interoduction of no synthase on renal toxicity of gentamicin were investegated. materials & methods: isolated kidneys of male rats were perfused with tyrode buffer (t) as the control group, t+l-argrinine as group 2, t+l-name as group 3, t+gentamicin (g) as group 4, t+l-arginine + urinary enzymes, lactate dehydrogenace and alkaline phosphatase (markers of cell death) were measured. results: enzymes activity in group 2, 3 and 5 were not significantly different from the control group, in contrast groups 4, 6 and 5, 4 were significanthy different. group 6 showed significant increase and group 5 showed a significant decrease in the enzymes activity as compared with group 4. conclusion: this study suggests that no can protect kidney from gentamicin toxicity.

Effects of nitric oxide on gentamicin toxicity in isolated perfused rat kidneys.
BACKGROUND There have been many studies in recent years concerning the role of nitric oxide (NO) in acute renal failure (ARF). In this study, the effects of the inhibition or the induction of NO synthase (NOS) on gentamicin-induced ARF was investigated in isolated perfused rat kidneys. METHODS Kidneys from male Sprague-Dawley rats were perfused in situ for 90 min. Perfusion was conducted in t...
full textEffects of lipopolysaccharide-induced septic shock on rat isolated kidney, possible role of nitric oxide and protein kinase C pathways
Objective(s): Pathophysiology of sepsis-associated renal failure (one of the most common cause of death in intensive care units) had not been fully determined. The effect of nitric oxide and protein kinase C (PKC) pathways in isolated kidney of Lipopolysaccharide-treated (LPS) rats were investigated in this study. Materials and Methods: Vascular responsiveness to phenylephrine and acetylcholine...
full textThe Isolated Perfused Rat Kidney
1. In the isolated rat kidney, perfused at constant pressure with a medium free from renin substrate, addition of angiotensin I to the perfusate decreases renal 'plasma' flow. 2. A peptide inhibitor, SQ 20881, of converting enzyme reduces the vasoconstrictor effect of angiotensin I up to a maximum of 87%, the degree of inhibition being dose-dependent. 3. The molar ratio of equi-effective doses ...
full textGluconeogenesis in the isolated perfused rat kidney.
The perfused rat kidney was used for a study of gluconeogenesis. The kidney was completely isolated and perfused with Krebs-Henseleit bicarbonate buffer containing 5% albumin. Perfusion flow rate and pressure remained stable for 2 to 3 hours, although the glomerular filtration rate diminished with time. The fractional reabsorption of both water and sodium was 0.92 during the 1st hour of perfusi...
full textThe Effects of L-Citrulline Pretreatment on the Isometric Tension of the Isolated Perfused Rat Aorta
Background: The achievement of an effective compound for prevention/treatment of hypertension with fewer complications has been of interest in recent years. OBJECTIVES: This study aimed to examine the effects of L-citrulline pretreatment on the tension of isolated rat aortic tissues precontracted by different vasoconstrictors. METHODS: Sixteen male Wistar rats (300-350g) were randomly divided i...
full textEffects of microcystin-LR in isolated perfused rat kidney.
Microcystin is a hepatotoxic peptide which inhibits protein phosphatase types 1 and 2A. The objective of the present study was to evaluate the physiopathologic effects of microcystin-LR in isolated perfused rat kidney. Adult Wistar rats (N = 5) of both sexes (240-280 g) were utilized. Microcystin-LR (1 microg/ml) was perfused over a period of 120 min, during which samples of urine and perfusate...
