expression of leishmania major lmsti1 in yeast pichia pastoris
Authors
abstract
background: leishmania major lmsti1 is a conserved protein among different species of leishmania, and expressed in both amastigote and promastigote forms of l. major life cycle. it has previously been expressed in bacterial systems. m aterials and methods: to express lmsti1 in the methylotrophic yeast pichia pastoris ( p. pastoris ), the shuttle vector ppicza containing gene lmsti1 was constructed under the control of the aox1 promoter. the recombinant vector was electro-transformed into p. pastoris, and induced by 0.5% methanol in the buffered medium. the expression of the lmsti1 protein was visualized in the total soluble protein of p. pastoris by 12% sds-page, and further confirmed by western blotting with l.major -infected mouse sera and hrp-conjugated goat anti-mouse igg as the first and secondary antibodies, respectively. results : the expression level was 0.2% of total soluble proteins. conclusion: it might be possible to use this formulation as a whole yeast candidate vaccine against cutaneous leishmanization.

Expression of Gp63 Gene from NIH Strain of Leishmania major in Pichia pastoris
Leishmaniasis is a major infectious disease of considerable public health in more than 86 countries around the world. Several approaches toward vaccine development against this disease have been taken. Glycoprotein (gp63) is conserved among diverse species of Leishmania and has induced immunological responses in murine models. Therefore, this glycoprotein has been considered as a second generat...

full textExpression of recombinant proteins in the methylotrophic yeast Pichia pastoris.
Protein expression in the microbial eukaryotic host Pichia pastoris offers the possibility to generate high amounts of recombinant protein in a fast and easy to use expression system. As a single-celled microorganism P. pastoris is easy to manipulate and grows rapidly on inexpensive media at high cell densities. Being a eukaryote, P. pastoris is able to perform many of the post-translational mo...
full textHeterologous protein expression in the methylotrophic yeast Pichia pastoris.
During the past 15 years, the methylotrophic yeast Pichia pastoris has developed into a highly successful system for the production of a variety of heterologous proteins. The increasing popularity of this particular expression system can be attributed to several factors, most importantly: (1) the simplicity of techniques needed for the molecular genetic manipulation of P. pastoris and their sim...
full textHeterologous Expression of Bovine Prochymosin in Pichia pastoris GS115
Objectives: In present research we evaluate the expression of this critical enzyme in a eukaryotic system for future use in cheese industry. Materials and Methods: We have cloned bovine prochymosin gene in methylotrophic yeast, P. pastoris, using pPIC9K as an expression vector. The recombinant plasmid was transformed into the host by electroporation, and it was expre...
full textEvaluation of pH/buffering conditions effect on the optimization of Recombinant Human Erythropoietin expression in the methylotrophic yeast, Pichia pastoris
Expression of recombinant proteins and drugs in Pichia pastoris has been in development since the late 1980s and the number of recombinant proteins produced in P. pastoris has increased significantly in the past several years. Unlike bacteria, this strain is capable of producing complex proteins with post translational modifications such as correct folding, glycosylation, proteolytic maturation...
