Do ∆F508 heterozygotes have a selective advantage?

چکیده
In this paper the fitness of the ∆F508 heterozygote is assessed and the age of the ∆F508 mutation in the cystic fibrosis locus is estimated. Data from three microsatellite loci are applied. The analysis is performed conditional on the present-day frequency of the ∆F508 mutation and based on assumptions about the demographic history of the European population and the mutation rate in the three microsatellite loci. It is shown that the data gives evidence of positive selection (up to 2–3% per ∆F508 heterozygote), but also that data could be explained by negative selection of roughly the same order of magnitude. The age of the ∆F508 mutation is subsequently estimated; it is found that the mutation is at least 580 generations old, but could be much older depending on the microsatellite mutation rate and the exact number of substitutions experienced in the history of the three microsatellite loci.

Relationship between genotype and phenotype for the CFTR gene W846X mutation.
Cystic fibrosis (CF) is the most common inherited disorder in white populations. It occurs in approximately 1/2500 live births and is characterised by chronic and progressive obstructive lung disease, pancreatic insufficiency, and high sweat electrolyte levels. Western Brittany has one of the highest rates of CF in the world. 2 Over 98% of the CF mutations in the Celtic population of Brittany h...
متن کاملEffects of a locus affecting floral pigmentation in Ipomoea purpurea on female fitness components.
A locus influencing floral pigment intensity in the morning glory, Ipomoea purpurea, is polymorphic throughout the southeastern United States. Previous work has suggested that the white allele at this locus has a transmission advantage during mating because of the effect of flower color on pollinator behavior. The experiment described here was designed to determine whether other effects of the ...
متن کاملDo ΔF508 Heterozygotes Have A Selective Advantage Against Influenza Virus?
The ΔF508 CFTR mutation, which is the commonest cause of cystic fibrosis (CF), has the highest frequency of any genetic mutation among Caucasians of central European origin. Persistence of a mutation suggests that heterozygosity for this mutation imparts some genetic advantage. We hypothesized that heterozygosity provides a selective survival advantage during influenza pneumonitis, as a result ...
متن کاملMHC heterozygosity confers a selective advantage against multiple-strain infections.
Genetic heterozygosity is thought to enhance resistance of hosts to infectious diseases, but few tests of this idea exist. In particular, heterozygosity at the MHC, the highly polymorphic loci that control immunological recognition of pathogens, is suspected to confer a selective advantage by enhancing resistance to infectious diseases (the "heterozygote advantage" hypothesis). To test this hyp...

1. Measurements of resting metabolic rate were made by open-circuit indirect calorimetry in 78 unrelated cystic fibrosis patients and 30 healthy control subjects. The aims of this study were: (i) to determine the range of variability in resting metabolic rate in cystic fibrosis, (ii) to relate this to pulmonary function and body size, and (iii) to investigate the hypothesis that, in cystic fibr...
