The small heat-shock protein αB-crystallin uses different mechanisms of chaperone action to prevent the amorphous versus fibrillar aggregation of α-lactalbumin.

چکیده
Stress conditions can destabilize proteins, promoting them to unfold and adopt intermediately folded states. Partially folded protein intermediates are unstable and prone to aggregation down off-folding pathways leading to the formation of either amorphous or amyloid fibril aggregates. The sHsp (small heat-shock protein) αB-crystallin acts as a molecular chaperone to prevent both amorphous and fibrillar protein aggregation; however, the precise molecular mechanisms behind its chaperone action are incompletely understood. To investigate whether the chaperone activity of αB-crystallin is dependent upon the form of aggregation (amorphous compared with fibrillar), bovine α-lactalbumin was developed as a model target protein that could be induced to aggregate down either off-folding pathway using comparable buffer conditions. Thus when α-lactalbumin was reduced it aggregated amorphously, whereas a reduced and carboxymethylated form aggregated to form amyloid fibrils. Using this model, αB-crystallin was shown to be a more efficient chaperone against amorphously aggregating α-lactalbumin than when it aggregated to form fibrils. Moreover, αB-crystallin forms high molecular mass complexes with α-lactalbumin to prevent its amorphous aggregation, but prevents fibril formation via weak transient interactions. Thus, the conformational stability of the protein intermediate, which is a precursor to aggregation, plays a critical role in modulating the chaperone mechanism of αB-crystallin.

Structural and functional aspects of hetero-oligomers formed by the small heat shock proteins αB-crystallin and HSP27.
BACKGROUND αB-crystallin and HSP27 are mammalian intracellular small heat shock proteins. RESULTS These proteins exchange subunits in a rapid and temperature-dependent manner. CONCLUSION This facile subunit exchange suggests that differential expression could be used by the cell to regulate the response to stress. SIGNIFICANCE A robust technique defines parameters for the dynamic interact...
متن کاملEffect of αB-Crystallin on Protein Aggregation in Drosophila
Disorganisation and aggregation of proteins containing expanded polyglutamine (polyQ) repeats, or ectopic expression of α-synuclein, underlie neurodegenerative diseases including Alzheimer's, Parkinson, Huntington, Creutzfeldt diseases. Small heat-shock proteins, such as αB-crystallin, act as chaperones to prevent protein aggregation and play a key role in the prevention of such protein disorga...
متن کاملThe Effect of Hoffmeister Salts on the Chaperoning Action of β-Casein in Preventing Aggregation of Reduced β-Lactalbumin
Protein aggregation and precipitation is associated with many debilitating diseases including Alzheimer's, Parkinson's, and light-chain amyloidosis. β-Casein, a member of the casein family, has been demonstrated to exhibit chaperone-like activity to protect protein form aggregation. Hofmeister salts (lyotropice series) are a class of ions which have an effect on the solubility and also the stab...
متن کاملReduced survival of lens epithelial cells in the α A - crystallin - knockout mouse
α-Crystallin is the major component of vertebrate eye lenses and constitutes approximately a third of the lens fiber cell proteins (Horwitz, 2000). It is also expressed at lower levels in lens epithelial cells. α-Crystallin is a hetero-oligomeric complex of two closely related polypeptides, αA and αB, present in 3:1 stoichiometry in lens fibers. The two subunits have chaperone activity, and are...
متن کاملR120G αB-crystallin promotes the unfolding of reduced α-lactalbumin and is inherently unstable
α-Crystallin is the principal lens protein which, in addition to its structural role, also acts as a molecular chaperone, to prevent aggregation and precipitation of other lens proteins. One of its two subunits, αBcrystallin, is also expressed in many non-lenticular tissues, and a natural missense mutation, R120G, has been associated with cataract and desminrelated myopathy, a disorder of skele...
