1Plant Breeding Institute, ARC, SF-31600 Jokioinen, Finland; 2Department of Genetics, SF-00014 University of Helsinki, Helsinki, Finland In a recent article describing the effects of various reaction conditions, Blanchard et al. ~1~ ment ioned that spermine and spermidine at concentrations between 0.5 and 3 mM had no effect on PCR amplification ~2'3> with their sequence-specific primers. Because we have observed with arbitrary primers and other DNA templates the concentrat ion-dependent effects of spermine and spermidine either alone or together in PCR, we would like to report our experiences here. The present experiments were run in 1991 and may therefore carry outdated features. The observation that barley (Hordeum vulgare) DNA samples isolated by two different methods, that of Wienand and Feix ~4~ and the other of Wagner et al., ~s) yielded different results when used as PCR templates gave an impetus to the present study. We deduced that a component was carried along with DNA from the solutions used in the latter method, which keeps the nucleic acids precipitated in the early stages of the procedure with PEG in the presence of polyamines. It appeared that the polyamines, spermine and spermidine, either alone or

The Effect of Different Polyamines on Some Physiological Traits as ACC Oxidase and Superoxide Dismutase Enzymes Activity in Chrysanthemum morifolium cv. ‘Bright Golden Ann’
This experiment was conducted to determine the effect of foliar spray with polyamines on postharvest life of chrysanthemum cut flowers in the horticulture laboratory of Islamic Azad University of Rafsanjan in 2015. For this purpose, an experiment was conducted based on a completely randomized design with three replications with spermidine, spermine and putrescine treatments at 1, 2 and 3 mM con...

متن کاملCharacterization of a novel spermidine/spermine acetyltransferase, BltD, from Bacillus subtilis.
Overexpression of the BltD gene in Bacillus subtilis causes acetylation of the polyamines spermidine and spermine. BltD is co-regulated with another gene, Blt, which encodes a multidrug export protein whose overexpression facilitates spermidine export [Woolridge, Vazquez-Laslop, Markham, Chevalier, Gerner and Neyfakh (1997) J. Biol. Chem. 272, 8864-8866]. Here we show that BltD acetylates both ...

In order to investigate the seasonal changes and the possible role of the free polyamines in the leaves and stems on the alternate bearing habit of the ‘Kinnow’ mandarin (Citrus reticulata Blanco) trees, a comparative study was conducted to analyze the levels of free polyamines (putrescine, spermidine and spermine) in the leaves and stems tissues of “on” and &ldquo...
متن کاملSpermidine but not spermine is essential for hypusine biosynthesis and growth in Saccharomyces cerevisiae: spermine is converted to spermidine in vivo by the FMS1-amine oxidase.
In our earlier work we showed that either spermidine or spermine could support the growth of spe2Delta or spe3Delta polyamine-requiring mutants, but it was unclear whether the cells had a specific requirement for either of these amines. In the current work, we demonstrate that spermidine is specifically required for the growth of Saccharomyces cerevisiae. We were able to show this specificity b...
