Total inhibition of hepatic beta-lipoprotein production in the rat by orotic acid.

چکیده
Lipoproteins of rat plasma have been characterized by lipid analysis, preparative ultracentrifugation, electrophoresis on paper, and immunoelectrophoresis with specific antisera to Lu-lipoprotein, P-lipoprotein, and whole rat plasma. Similar techniques were used to characterize the lipoproteins released by livers perfused in situ with a suspension of erythrocytes in a medium initially free of rat plasma proteins. Normal rat livers released a-lipoprotein, P-lipoprotein, albumin, and other plasma proteins during the perfusion. When rats were fed erotic acid as 1% of their diet, the plasma @lipoprotein concentration fell within 7 days to less than 1% of normal. It rebounded to normal within 48 hours following withdrawal of erotic acid or supplementation of the diet with 0.17% of adenine. When perfused in situ, livers from erotic acid-fed rats released cr-lipoprotein, albumin, and other plasma proteins but no detectable /?-lipoprotein. They released smaller amounts of cholesterol and phospholipid than normal livers and no triglyceride, although they contained 10 times normal amounts of triglycerides. The fatty liver produced in rats by erotic acid appears to result from an inhibition of synthesis or release of hepatic @-lipoprotein, supporting the concept that ,&lipoprotein has a specific role in the normal transport of triglyceride out of the liver.

Radioimmunoassay of beta lipoprotein-protein of rat serum.
A double antibody radioimmunoassay for rat serum beta lipoprotein-protein (beta Lp-protein) is described. The protein was purified by ultracentrifugation, selective heparin-manganous precipitation, and gel filtration on Sephadex G-200. Antiserum was prepared in rabbits by biweekly immunization and absorbed with nonbeta lipoprotein containing rat serum. Iodination with (125)I and purification by...

متن کاملHypolipemic action of glucagon in experimental endogenous lipemia in the rat.
The effect of glucagon on serum lipids and very low density lipoproteins (VLDL) has been examined in the eulipemic and the hyperlipemic rat. An inhibition of amino acid incorporation into hepatic lipoprotein apoprotein was observed, with an associated decrease in circulating VLDL apoprotein, decreased serum triglyceride concentration, and a loss of the pre-beta band as judged by serum lipoprote...
متن کاملFenofibrate prevents orotic acid--induced hepatic steatosis in rats.
The experiments performed in this report were designed to investigate the mechanisms involved in the metabolic alterations associated with orotic acid-induced hepatic steatosis and the effect of fenofibrate, a stimulant of peroxisome proliferators-activated receptor alpha (PPARalpha), on these alterations. Male Wistar rats were divided into three experimental groups: 1) fed a balanced diet (C);...
متن کاملApolipoproteins of the orotic acid fatty liver: implications for the biogenesis of plasma lipoproteins.
Rats fed orotic acid develop fatty livers characterized by triglyceride-laden, membrane-bounded vesicles designated "liposomes." We have measured the levels of apolipoproteins in isolated liposomes and other subcellular fractions by SDS-polyacrylamide gel electrophoresis, electrotransfer, and immunodecoration. Apolipoproteins Bh, Bl, E, and C appear to cofractionate; for these proteins, the lip...
