چکیده

Chapter 1. Magnesium-dependent phosphate ester processing enzymes frequently employ a carboxylate-bridged dimetallic unit in their active sites. Biomimetic modelling chemistry of these magnesium centers has not been investigated previously. An brief introduction of the enzyme systems is presented, along with the focus of the present work. Chapter 2. Several dimagnesium(II) complexes were synthesized and characterized by employing the dinucleating ligand XDK, where H2XDK is m-xylylenediamine bis(Kemp's triacid imide), to stabilize the {Mg(4t-O2CR)}2 2+ unit. A [Mg2(XDK)(CH 3OH)4(H20) 2(NO 3)] complex has been prepared and allowed to react with one or two equivalents of diphenyl phosphate (DPP) to yield [Mg 2 (XDK)(DPP)(CH 3OH)3(H20)(NO3)] and [Mg2(XDK)(DPP)2(CH 3OH)3(H20)], respectively. These are the first structurally characterized phosphate ester-bridged dimagnesium complexes which are potential phosphatase models. Chapter 3. The phosphate ester exchange rate of free and bound diphenyl phosphate in [Mg 2(XDK)(DPP)2(CH 3OH)3(H20)] in methanol has been measured by variable temperature 3 1P{ 1H} NMR spectroscopy and compared to that of structurally analogous dizinc(II) and dicalcium(II) complexes. The synthesis and structural characterization of the dicalcium(II) complex is presented, along with a discussion on the differences between the carboxylateand phosphate ester-bridged dimagnesium, dizinc, and dicalcium centers. Chapter 4. Modelling studies of magnesium-dependent phosphatase enzymes were extended into aqueous media with the synthesis of a water-soluble XDK analog, WXDK. The synthesis and characterization of the carboxylate-bridged dimagnesium-WXDK complexes are described. The biological relevance of these modelling studies is discussed. Thesis Supervisor: Stephen J. Succinic Acid Monoethyl Ester and Metformin Regulates Carbohydrate Metabolic Enzymes and Improves Glycemic Control in Streptozotocin-Nicotinamide Induced Type2 Diabetic Rats

Objective. Succnic acid mono ethyl ester (EMS) was recently proposed as an insulinotropic agent for the treatment of non-insulin dependent diabetes mellitus. In the present study the effect of EMS and Metformin on the activities of carbohydrate metabolic enzymes in streptozotocinnicotinamide induced type 2 diabeteic model was investigated. Methods. EMS were injected intraperitonially at doses 2...

THE FORMATION OF GLUCOSE-l-PHOSPHORIC ACID IN EXTRACTS OF MAMMALIAN TISSUES AND OF YEAST

The initial stages of fermentation of glycogen to lactic acid in muscle have been investigated only recently. It was shown (1, 2) that the glycolytic reactions are initiated by the formation of glucose-l-phosphoric acid from glycogen and inorganic phosphate, an enzymatic process in which adenylic acid acts as coenzyme. The glucose-l-phosphoric acid (l-ester) is converted to glucose-6phosphoric ...

Gene expression and control of enzymes for synthesis of magnesium protoporphyrin monomethyl ester in Rhodobacter sphaeroides.

The photosynthetic gene cluster of R sphaeroides is encoded on a 45kb region of the genome and is represented by 8 overlapping inserts cloned in the mobilisable vector pSUP202 [ l ] . Characterisation of the cluster by a variety of techniques, including complementation analyses and study of accumulated products of bacteriochlorophyll synthesis in mutants, provides a physical map (Figure 1) and ...

Regulation of chloroplast photosynthetic activity by exogenous magnesium.

Magnesium was most inhibitory to photosynthetic reactions by intact chloroplasts when the magnesium was added in the dark before illumination. Two millimolar MgCl(2), added in the dark, inhibited CO(2)-dependent O(2) evolution by Hordeum vulgare L. and Spinacia oleracea L. (C(3) plants) chloroplasts 70 to 100% and inhibited (pyruvate + oxaloacetate)-dependent O(2) evolution by Digitaria sanguin...
