چکیده

The primary cellular source of FVIII biosynthesis is controversial, with contradictory evidence supporting an endothelial or hepatocyte origin. LMAN1 is a cargo receptor in the early secretory pathway that is responsible for the efficient secretion of FV and FVIII to the plasma. Lman1 mutations result in combined deficiency of FV and FVIII, with levels of both factors reduced to ~10-15% of normal in human patients. We generated Lman1 conditional knockout mice to characterize the FVIII secretion profiles of endothelial cells and hepatocytes. We demonstrate that endothelial cells are the primary biosynthetic source of murine FVIII, and that hepatocytes make no significant contribution to the plasma FVIII pool. Utilizing RiboTag mice and polyribosome immunoprecipitation, we performed endothelial cell-specific mRNA isolation and qPCR analyses to confirm that endothelial cells highly express F8, and to explore the heterogeneity of F8 expression in different vascular beds. We demonstrate that endothelial cells from multiple, but not all, tissues contribute to the plasma FVIII pool in the mouse. Plenary Paper THROMBOSIS AND HEMOSTASIS Murine coagulation factor VIII is synthesized in endothelial cells

Plasma coagulation factor VIII (FVIII) is tightly associated with von Willebrand factor (VWF). Though it is well established that VWF is synthesized exclusively in endothelial cells andmegakaryocytes, the cellular source of FVIII has been debated for decades. The delayed rise of FVIII in von Willebrand disease patients infused with VWF demonstrates that VWF and FVIII do not require synthesis in...

HEMOSTASIS, THROMBOSIS, AND VASCULAR BIOLOGY Intrinsic pathway of blood coagulation contributes to thrombogenicity of atherosclerotic plaque

Thrombosis is the major mechanism underlying acute complications of atherosclerosis. Although thrombogenicity of atherosclerotic plaques has been ascribed to activation of the extrinsic pathway of blood coagulation, in the present study we investigated contribution of the intrinsic factor VIII (fVIII)–dependent pathway. We found that in vitro exposure of human macrophages and smooth muscle cell...

von Willebrand factor: a marker of endothelial damage?

Von Willebrand factor (vWF), a large glycoprotein encoded by a gene on chromosome 12, is synthesized by vascular endothelial cells and circulates in human plasma at concentrations of 10 mg/mL.* In plasma, vWF forms a noncovalent complex with coagulation factor VIII, the protein encoded by a gene on the X chromosome that is deficient or defective in hemophilia A. This molecular complex is essent...

Regular Article THROMBOSIS AND HEMOSTASIS A von Willebrand factor fragment containing the D9D3 domains is sufficient to stabilize coagulation factor VIII in mice

Loss of procoagulant factor VIII (FVIII) function results in pathologic bleeding.Noncovalent binding to vonWillebrand factor (VWF) protects FVIII from degradation and rapid clearance from plasma. FVIII levels in patients with vonWillebrand disease (VWD) type 1 or type 3 are generally reduced concomitant with decreased levels of plasma VWF. Patients with VWF mutations interfering with VWF: FVIII...

THROMBOSIS AND HEMOSTASIS Intracellular cotrafficking of factor VIII and von Willebrand factor type 2N variants to storage organelles

Weibel-Palade bodies (WPBs) are the endothelial storage organelles that are formed upon von Willebrand factor (VWF) expression. Apart from VWF, WPBs contain a variety of hemostatic and inflammatory proteins. Some of these are thought to be targeted to WPBs by directly interacting with VWF in the secretory pathway. Previous studies have demonstrated that coexpression of factor VIII (FVIII) with ...
