چکیده

GOLDSTEIN, LEON. Relation of glutamate to ammonia producfion in the rat kidney. Am. J. Physidl. 210(3): 661-666. Ig66.-Glutamate was an effective inhibitor of isolated rat renal glutaminase at concentrations normally found in the kidney. Injection of monosodium glutamate into acidotic rats inhibited renal ammonia excretion about 507~ with no significant effect on urinary PH. Alterations in acid-base balance known to increase (metabolic acidosis) and decrease (metabolic alkalosis) renal ammonia production caused significant changes in renal glutamate concentrations. Metabolic acidosis (40 mmoles NH&l/kg per day) produced a rapid (I day) and sustained (3 days) decrease (35 TO) in glutamate concentrations. Injection of acetazolamide (300 mg/kg per day) reduced glutamate concentration 20% within 3 days. Metabolic alkalosis (40 mmoles NaHC03/kg per day) produced a gradual increase (25%) in glutamate concentration over a period of 3 days. These results suggest that glutamate may play a role in the regulation of renal ammonia production.

Renal gluconeogenesis in acidosis, alkalosis, and potassium deficiency: its possible role in regulation of renal ammonia production.

In man, dog, and rat the induction of metabolic acidosis leads to an increase in renal production and excretion of ammonia (2-4) that appears to derive primarily from glutamine (4-6). Studies of the renal cortex of the acidotic rat reveal an increase in the activity of glutaminase I, the enzyme that deamidates glutamine to glutamate (Figure 1), and it is assumed that this may contribute to the ...

Hepatic Glutamate Dehydrogenase Activity and the Presence of Ammonia and Urea in the Circulatory Fluid of Channa Gachua in Water-Restricted Condition

Ammonia is the chief excretory product in fishes. However, non-availability of enough of water in the habitat, may lead to the formation of urea, in fishes. In the present study, the possible role of urea formation to avoid the toxicity of ammonia under water-restricted condition was tested in Channa gachua. Circulatory urea and ammonia were estimated in the blood of the fishes and glu...

Glutamate dehydrogenase activation and ammonia formation by rat kidney mitochondria.

Mitochondria from rat renal cortex were incubated with physiological concentrations of glutamate, or glutamine, or both. Changes in medium ammonia, glutamate, glutamine, and aspartate were measured and fluxes through glutamate dehydrogenase, glutaminase, and glutamate oxalacetate transaminase were calculated at different pH levels in vitro and in mitochondria from animals in which metabolic aci...

Effect of Pimpinella tirupatiensis on Oxidative Enzymes in STZ-induced Diabetic Rat Kidney

The present study was aimed to evaluate the therapeutic potential of Pimpinella tirupatiensis (Pt) by assaying the activities of selective mitochondrial enzymes in streptozotocin induced diabetic rats. Diabetic rats showed a significant (p < 0.01) reduction in the activities of oxidative enzymes Succinate dehydrogenase (SDH), Malate dehydrogenase (MDH), Glutamate dehydrogenase (GDH) and isocitr...
