Polyamines and the delay in deoxyribonucleic acid synthesis in some bacteriophage T4 infections.

چکیده
Escherichia coli B infected by the DD mutant of T4, am N116, is stimulated to initiate deoxyribonucleic acid accumulation by 1 to 10 mm spermidine but not by 10 mm putrescine. The syntheses of putrescine and spermidine in cells infected by T4D and the mutant are similar, although slight differences are observed in the intracellular concentration of free spermidine. Unlike r-K12 (lambda) systems, am N116-infected cells do not leak polyamine.

Polyamines in the synthesis of bacteriophage deoxyribonucleic acid. II. Requirement for polyamines in T4 infection of a polyamine auxotroph.
Polyamine depletion produced by exogenous arginine in Escherichia coliK-12 cultures defective in agmatine ureohydrolase activity resulted in a marked inhibition of the rates of growth and nucleic acid synthesis. Addition of putrescine or spermidine to such depleted cultures restored the control rate of growth and nucleic acid accumulation. The omission of lysine resulted in a further decrease i...
متن کاملThe role of polyamines in the neutralization of bacteriophage deoxyribonucleic acid.
It was previously reported that bacteriophage T4 contains the polyamines putrescine, NH2(CH2)$;IH2, and spermidine, NHQ(CH&NH(CH&NH2, in amounts sufficient to neutralize about half of the viral deoxyribonucleic acid (1). The putrescine and spermidine in the phage were found to be derived from the large amount of these polyamines normally present in the host bacterium, Escherichia coli B. It was...
متن کاملInhibition of host deoxyribonucleic acid synthesis by T4 bacteriophage in the absence of protein synthesis.
The requirement for phage protein synthesis for the inhibition of host deoxyribonucleic acid synthesis has been investigated by using a phage mutant unable to catalyze the production of any phage deoxyribonucleic acid. It has been concluded that the major pathway whereby phage inhibit host syntheses requires protein synthesis. The inhibition of host syntheses by phage ghosts is not affected by ...
متن کاملEffect of hydroxyurea on replication of bacteriophage T4 in Escherichia coli.
Hydroxyurea inhibited the replication of bacteriophage T4 in Escherichia coli B. The concentration of hydroxyurea required to inhibit net deoxyribonucleic acid (DNA) synthesis 50% was about 50-fold less than that required in uninfected cells. Even in the presence of high hydroxyurea concentrations, phage DNA was readily synthesized from the products of breakdown of the E. coli DNA, and viable p...
متن کاملThe Effect of Chloramphenicol on Deoxyribonucleic Acid Synthesis and the Development of Resistance to Ultraviolet Irradiation in E. Coli Infected with Bacteriophage T2
To elucidate the role of protein synthesis in DNA formation, E. coli R2 infected with phage T2 was studied as a model, employing chloramphenicol to inhibit protein synthesis. The following results were obtained. 1. Chloramphenicol inhibited protein synthesis but not synthesis of nucleic acids in uninfected bacteria. 2. Studies of the effect of chloramphenicol on phage maturation indicated a del...
