چکیده

Protein translocation in cells has been modelled by Brownian ratchets. In such models, the protein diffuses through the nanopore by thermal fluctuations. On one side of the pore ratcheting molecules bind to the protein and hinder it to diffuse out of the pore. We study a simple Brownian ratchet by means of a reflected Brownian motion (Xt)t≥0 with a changing reflection point (Rt)t≥0. The rate of change of Rt is γ(Xt − Rt) and is distributed uniformly on [Rt;Xt]. We show that the asymptotic speed of the ratchet scales with γ and the asymptotic variance is independent of γ.

The brownian ratchet and power stroke models for posttranslational protein translocation into the endoplasmic reticulum.

A quantitative analysis of experimental data for posttranslational translocation into the endoplasmic reticulum is performed. This analysis reveals that translocation involves a single rate-limiting step, which is postulated to be the release of the signal sequence from the translocation channel. Next, the Brownian ratchet and power stroke models of translocation are compared against the data. ...

Charge requirements for proton gradient-driven translocation of anthrax toxin.

Anthrax lethal toxin is used as a model system to study protein translocation. The toxin is composed of a translocase channel, called protective antigen (PA), and an enzyme, called lethal factor (LF). A proton gradient (ΔpH) can drive LF unfolding and translocation through PA channels; however, the mechanism of ΔpH-mediated force generation, substrate unfolding, and establishment of directional...

Cellular motions and thermal fluctuations: the Brownian ratchet.

We present here a model for how chemical reactions generate protrusive forces by rectifying Brownian motion. This sort of energy transduction drives a number of intracellular processes, including filopodial protrusion, propulsion of the bacterium Listeria, and protein translocation.

Two-way communication between SecY and SecA suggests a Brownian ratchet mechanism for protein translocation

The essential process of protein secretion is achieved by the ubiquitous Sec machinery. In prokaryotes, the drive for translocation comes from ATP hydrolysis by the cytosolic motor-protein SecA, in concert with the proton motive force (PMF). However, the mechanism through which ATP hydrolysis by SecA is coupled to directional movement through SecYEG is unclear. Here, we combine all-atom molecul...

Realistic extensions of a Brownian ratchet for protein translocation

We study a model for the translocation of proteins across membranes through a nanopore using a ratcheting mechanism. When the protein enters the nanopore it diffuses in and out of the pore according to a Brownian motion. Moreover, it is bound by ratcheting molecules which hinder the diffusion of the protein out of the nanopore, i.e. the Brownian motion is reflected such that no ratcheting molec...
