Neurotoxins from Wecffewys Spider Venom are Potent Presynaptic Blockers in Drosophila
نویسندگان
چکیده
Studies of presynaptic events in synaptic transmission may be facilitated through the use of specific ligands for functional components of the transmitter release mechanism and through the use of genetics. For this purpose, neurotoxins that affect neuromuscular transmission in Drosophila have been identified and purified from Plecfreurys spider venom (PLTX). One class of toxins causes irreversible presynaptic block, probably by blocking calcium entry or by acting on other closely associated processes. These toxins have been highly purified and are peptides of about 7 kDa in molecular weight. They specifically block transmitter release at nanomolar concentrations and may be useful in further biochemical studies.

Identification and purification of an irreversible presynaptic neurotoxin from the venom of the spider Hololena curta.
A search for potent toxins that inhibit neuronal calcium channels in Drosophila melanogaster has resulted in the identification of a presynaptic neurotoxin from the venom of the hunting spider Hololena curta. Using Drosophila neuromuscular junction as an assay, presynaptic inhibitory activity was purified using gel filtration and reverse-phase HPLC. Data from gel electrophoresis indicate that t...

Besides the power of the chelicerae, synergistic interactions between different components in the venom of Cupiennius salei ensure the hunting success of this spider. The main components of the venom were tested alone or in combination according to their physiological venom concentrations in Drosophila bioassays. The high K+ ion content of the venom synergistically increases the insecticidal ac...
متن کاملP 25: The Facilitatory Action of Snake Venom Phospholipase A2 Neurotoxins by Which Increase the Release of Acetylcholine, May Improve Alzheimer\'s Disease Symptoms
Introduction: In a serious brain disorder like Alzheimer's disease, the levels of acetylcholine (Ach) drop significantly. The gradual death of cholinergic brain cells leads to a profound loss of memory and learning ability. Acetylcholine is the chemical messenger that sends messages from one neuron to another in the area of the brain used for memory. Many of the current medications act to enhan...
متن کاملCyto-insectotoxins, a novel class of cytolytic and insecticidal peptides from spider venom.
Eight linear cationic peptides with cytolytic and insecticidal activity, designated cyto-insectotoxins (CITs), were identified in Lachesana tarabaevi spider venom. The peptides showed antibiotic activity towards Gram-positive and Gram-negative bacteria at micromolar concentrations as well as toxicity to insects. The primary structures of the toxins were established by direct Edman sequencing in...
متن کاملalpha-Latrotoxin and its receptors: neurexins and CIRL/latrophilins.
alpha-Latrotoxin, a potent neurotoxin from black widow spider venom, triggers synaptic vesicle exocytosis from presynaptic nerve terminals. alpha-Latrotoxin is a large protein toxin (120 kDa) that contains 22 ankyrin repeats. In stimulating exocytosis, alpha-latrotoxin binds to two distinct families of neuronal cell-surface receptors, neurexins and CLs (Cirl/latrophilins), which probably have a...
