چکیده

Following infection, the development of phage P22 by either the lytic or the lysogenic pathways requires recombination, mediated either by the phage erf system or by the bacterial ret system [Botstein, D., and Matz, M. J. (1970) J. Mol. Biol. 54, 417-4401. We have investigated the timing of the essential recombinational processes with temperature-shift experiments using a temperature-sensitive erf mutant. In retcells, erf function appears to be required early in the infection to complete some essential step, the timing of which is the same in both the lytic and lysogenic circumstances. Once the step has taken place, subsequent development can occur without further erf function. However, the bulk of recombinant progeny arising in lytic crosses in retcells result from nonessential erf action late in the infection, after the time of the required early step.

Recombinational circularization of Salmonella phage P22 DNA.

The development of phage P22 following infection involves a mandatory recombination step, the timing of which is the same in both the lytic and lysogenic pathways. Covalently circular molecules of phage DNA were identified by alkaline sucrose sedimentation of infected cell lysates. The time of appearance of these structures corresponds to the time of the essential recombination step. On infecti...

Generalized transduction by phage P22 in Salmonella typhimurium. I. Molecular origin of transducing DNA.

P22, a temperate phage which grows on Salmoneua typhimurium, is capable of carrying out general&d transduction in this host. The general&d transducing particles are the same size ss P22 phage particles but instead of phage DNA they contain bacterial DNA which was synthesized before phage infection. Further, DNA molecules isolated from the transducing particles have the same molecular weight as ...

A recombination function essential to the growth of bacteriophage P22.

Mutants of the Sahrwneliu phage P22 have been isolated which cannot grow after infection of recombination-deficient (ret-) hosts. These mutants (designated erffor essential recombination function) are defective in a phage-specified recombination system, as shown by twoand three-factor crosses carried out in WC + and rethosts. Four erf mutants examined in detail fall into one complementation gro...

Transduction by phage P22 in a recombination-deficient mutant of Salmonella typhimurium.

Recombination-deficient (Ret-) mutants of Salmonella typhitn&um were used to study special transduction by phage P22 in the absence of the usual background of general transduction. Retstrains could not be transduced for any marker with a lysate arising by infection. When the same strains were infected with a lysate obtained by induction, low but significant levels of transduction were observed ...

Specialized transduction of tetracycline resistance by phage P22 in Salmonella typhimurium. II. Properties of a high-frequency-transducing lysate.

A high-frequency-transducing (HFT) lysate for tetracycline resistance (Lets) was obtained by inducing an unusual tets P22 lysogen which had been made by transducing Salmonella typhimurium LT.2 to letR with P22 phage grown on an LT-2 strain carrying the R factor 222. The HFT lysate contains defective P22 particles, called P22 Tc-10, which cannot grow or lysogenize (i.e., transduce tetn) upon sin...
