A Machine Learning Model to Predict the Onset of Alzheimer Disease using Potential Cerebrospinal Fluid (CSF) Biomarkers
نویسنده
چکیده
Clinical studies in the past have shown that the pathology of Alzheimer’s disease (AD) initiates, 10 to 15 years before the visible clinical symptoms of cognitive impairment starts to appear in AD diagnosed patients. Therefore, early diagnosis of the AD using potential early stage cerebrospinal fluid (CSF) biomarkers will be valuable in designing a clinical trial and proper care of AD patients. Therefore, the goal of our study was to generate a classification model to predict earlier stages of the AD using specific early-stage CSF biomarkers obtained from a clinical Alzheimer dataset. The dataset was segmented into variable sizes and classification models based on three machine learning (ML) algorithms, such as Sequential Minimal Optimization (SMO), Naïve Bayes (NB), and J48 were generated. The efficacy of the models to accurately predict the cognitive impairment status was evaluated and compared using various model performance parameters available in Weka software tool. The current findings show that J48 based classification model can be effectively employed for classifying cognitive impaired Alzheimer patient from normal healthy individuals with an accuracy of 98.82%, area under curve (AUC) value of 0.992 and sensitivity & specificity of 99.19% and 97.87%, respectively. The sample size (60% training and 40% independent test data) showed significant improvement in T-test with J48 algorithm when compared with other classifiers tested on Alzheimer dataset. P 62: Markers of Neuroinflammation Related to Alzheimer\'s Disease Pathology in the Elderly
Alzheimer Disease (AD) is a neurodegenerative disorder and the most common form of dementia. Increasing evidence suggests that Alzheimer's disease pathogenesis is not restricted to the neuronal compartment, but includes strong interactions with immunological mechanisms in the brain. In vitro and animal studies have linked neuroinflammation to Alzheimer's disease (AD) pathology. Studies on marke...
متن کاملThe most valuable biomarkers of Alzheimer\'s disease: a review article
Alzheimer’s disease (AD) is the most popular type of dementia in elderly and is described by a progressive loss of cognitive capacity and severe neurodegeneration which typically begins with memory deficits. The major biomarkers of AD include total tau, phosphorylated-tau and 42 amino acid isoform of amyloid beta that reflect neurodegeneration and indicate the pathophysiological processes in AD...
متن کاملMultiplexed Immunoassay Panel Identifies Novel CSF Biomarkers for Alzheimer's Disease Diagnosis and Prognosis
BACKGROUND Clinicopathological studies suggest that Alzheimer's disease (AD) pathology begins ∼10-15 years before the resulting cognitive impairment draws medical attention. Biomarkers that can detect AD pathology in its early stages and predict dementia onset would, therefore, be invaluable for patient care and efficient clinical trial design. We utilized a targeted proteomics approach to disc...
متن کاملCerebrospinal Fluid Indices in Acute Drug Intoxication Do they Predict the Patients’ Outcome?
Introduction: In some intoxicated patients, cerebrospinal fluid (CSF) is examined due to the prolonged loss of consciousness, focal neurologic findings, and fever of unknown origin. We aimed to evaluate the probable relationship between the different toxicity causes and the CSF indices in poisoned patients and determine if they could predict the patients’ outcome. Methods: All patients who h...
متن کاملCorrelation of cerebrospinal fluid levels of tau protein phosphorylated at threonine 231 with rates of hippocampal atrophy in Alzheimer disease.
BACKGROUND The microtubule-associated tau protein abnormally phosphorylated at threonine 231 (p-tau231) has been investigated as a potential marker of Alzheimer disease. Levels of cerebrospinal fluid (CSF) p-tau231 vary across patients with Alzheimer disease. We hypothesized that these variations partially reflect differences in the degree of neuronal damage and therefore may be used to predict...
