Inhibition of DNA and RNA synthesis by daunorubicin in sensitive and resistant Ehrlich ascites tumor cells in vitro.

چکیده
The effect of daunorubicin (DNR) on nucleic acid synthesis was investigated in vitro in cells from an Ehrlich ascites tumor that was sensitive in vivo to DNR and in cells from a subline of the tumor in which resistance to DNR had been developed in vivo. DNR inhibited DNA and RNA synthesis in sensitive as well as in resistant cells, as determined from the incorporation of three different radioactive precursors. In order to give the same degree of inhibition of DNA and RNA synthesis in resistant cells as in sensitive cells, the DNR concentrations required were approximately 5 times greater. When the synthesis of different RNA species in sensitive cells was analyzed, nucleoplasmic heterodisperse RNA was found to be the least DNR sensitive, while tRNA, 5 S RNA, and two small-molecular-weight RNA components had a medium sensitivity to DNR. Labeling of rRNA and of the 45 S and 32 S nucleolar RNA was found to be inhibited most and to the same extent, indicating that it is the synthesis of 45 S RNA that is inhibited and not the processing of 45 S RNA to rRNA. The preferential inhibition of rRNA was less pronounced in resistant cells than in sensitive cells, thus indicating that the sensitivity ofrRNA synthesis was changed more Muring development of resistance than was the sensitivity of synthesis of other RNA components. The cellular uptake of DNR was less in resistant than in sensitive cells, but the difference was too small to explain the differences in inhibition of the nucleic acid synthesis.

متن کاملVerapamil restoration of daunorubicin responsiveness in daunorubicin-resistant Ehrlich ascites carcinoma.
We have studied the influence of verapamil hydrochloride on the in vitro and in vivo effects of daunorubicin in Ehrlich ascites carcinoma. Daunorubicin-sensitive tumor was rendered resistant to daunorubicin by the continuous treatment of sequential generations of tumor-bearing BALB/c mice. The ability of daunorubicin to inhibit [(3)H]uridine and [(3)H]thymidine incorporation and the effect of d...
متن کاملInhibition of DNA and RNA Metabolism by Daunorubicin and Adriamycin in L1210 Mouse Leukemia1
Effects of daunorubicin and its new analog, adriamycin, on nucleic acid metabolism were studied in vitro in L1210 mouse leukemia cells with labeled nucleoside precursors and were compared with the established effects of actinomycin D. L1210 ascites tumor cells incubated with daunorubicin under physiological conditions of pH, temperature, and tonicity showed significantly greater inhibition of t...
متن کاملEnhancement by cyclosporin A of daunorubicin efficacy in Ehrlich ascites carcinoma and murine hepatoma 129.
Cyclosporin A abrogates pleiotropic drug resistance in certain experimental tumors. Its impact on drug-sensitive tumors has not been investigated. Our studies show that in drug-sensitive Ehrlich ascites carcinoma and hepatoma 129 cyclosporin A enhances daunorubicin inhibition of DNA synthesis in vitro and prolongs survival of host mice in vivo. Of particular interest is that cyclosporin A conve...
متن کاملSelective Inhibition by Sorbylhydroxamic Acid of Deoxyribonucleic Acid Synthesis in Ehrlich Ascites Tumor Cells1
Sorbylhydroxamic acid (SOHA) inhibited the incorporation in vitro of thymidine into deoxyribonucleic acid in Ehrlich ascites tumor cells, with no appreciable effect on the incorporation of uridine into ribonucleic acid or of L-leucine into protein. Hexanoylhydroxamic acid, devoid of the conjugated double-bond system, had no such activity. The concentration of SOHA conferring 50% inhibition of D...
