Assessing the Bacterial Ecology of Organohalide Respiration for the Design of Bioremediation Strategies

چکیده
Groundwater microbial community samples are traditionally collected using pumping techniques optimized for groundwater chemistry assessment although the impact of groundwater pumping parameters on apparent bacterial community structures (BCS) is not really known. We therefore studied the impact of pumping lift, flow regime and tubing material on BCS, which were analyzed by terminal-restriction fragment length polymorphism (T-RFLP). Ruzicka dissimilarity coefficients were calculated between T-RFLP profiles to assess disparities between BCS. Variations in pumping lift, flow regime and tubing material did not affect the apparent BCS in experiments using a homogenous water body under laboratory conditions showing that the conditions within the tube had no detectable effect on BCS. However, pumping groundwater from aquifer monitoring wells at different flow rates in the field revealed a significant impact on the apparent BCS. Water samples collected from fine sediment were the most affected by the pumping flow rate. Impact assessment of pumping parameters on groundwater bacterial communities ~ 33 ~ 2.

Organohalide respiration: microbes breathing chlorinated molecules.
Bacterial respiration has taken advantage of almost every redox couple present in the environment. The reduction of organohalide compounds to release the reduced halide ion drives energy production in organohalide respiring bacteria. This process is centred around the reductive dehalogenases, an iron-sulfur and corrinoid containing family of enzymes. These enzymes, transcriptional regulators an...
متن کاملOrganohalide Respiring Bacteria and Reductive Dehalogenases: Key Tools in Organohalide Bioremediation
Organohalides are recalcitrant pollutants that have been responsible for substantial contamination of soils and groundwater. Organohalide-respiring bacteria (ORB) provide a potential solution to remediate contaminated sites, through their ability to use organohalides as terminal electron acceptors to yield energy for growth (i.e., organohalide respiration). Ideally, this process results in non-...
متن کاملImpact of Ammonium on Syntrophic Organohalide-Respiring and Fermenting Microbial Communities
Syntrophic interactions between organohalide-respiring and fermentative microorganisms are critical for effective bioremediation of halogenated compounds. This work investigated the effect of ammonium concentration (up to 4 g liter(-1) NH4 (+)-N) on trichloroethene-reducing Dehalococcoides mccartyi and Geobacteraceae in microbial communities fed lactate and methanol. We found that production of...

Butachlor (BUT) is a chloroacetanilide herbicide widely applied to rice paddies to control annual grass and broad-leaf weeds. A BUT-degrading bacterial strain (PK) was isolated from paddy soils. Biochemical and 16S rRNA sequencing characteristics confirmed the strain as Pseudomonas aeruginosa (99% resemblance). The isolate dissipated BUT (100 μg/mL) in an M9 liquid medium with a rate of 0.5 ± 0...
