3-Arylcoumarin derivatives manifest anti-proliferative activity through Hsp90 inhibition.

چکیده
The potential therapeutic benefits associated with Hsp90 modulation for the treatment of cancer and neurodegenerative diseases highlight the importance of identifying novel Hsp90 scaffolds. KU-398, a novobiocin analogue, and silybin were recently identified as new Hsp90 inhibitors. Consequently, a library of 3-arylcoumarin derivatives that incorporated the structural features of KU-398 and silybin was designed, synthesized and evaluated against two breast cancer cell lines. Western blot analysis confirmed that the resulting 3-arylcoumarin hybrids target the Hsp90 protein folding machinery.

Heat Shock Protein 90 c-Terminal Inhibitors in Cancer Treatment
Coumarine (2H-1-benzopyran-2-ones) is benzopyrone class compounds and its derivatives are shown a wide spectrum of biological activity. Particularly, coumarine derivative compounds (novobiocin, coumermycin A1, KU135, and chlorobiocin) have an affinity for binding to the CTD and disrupting Hsp90 dimerization. Consequently, these compounds inhibit proper folding of oncogenic client proteins and c...

Heat shock protein 90 (Hsp90) is a molecular chaperone that is required for the maturation and activation of a number of client proteins, many of which are involved in cancer development. The ansamycin family of natural products and their derivatives, such as geldanamycin (GA), are well-known inhibitors of the essential ATPase activity of Hsp90. Despite structural studies on the complexes of an...

متن کاملInteraction Study of 1, 3 Substituted Isatin Derivatives with Anti Inflammatory Properties with Cyclooxygenase 1 and 2 Enzymes by Molecular Docking Method
Introduction: Inflammation as the body's defense response is accompanied with various diseases. Prostaglandins are major mediators of inflammation produced by the cyclooxygenase enzymes. So inhibitors of these enzymes can be effective in treating inflammation. There are reports of inhibition of these enzymes by isatin derivatives to control inflammation. Isatin is a heterocyclic compound whose...
