چکیده

immediate response of eukaryotic cells to oxidative. PARP-1 and PARP-2 display catalytic activity by contact with DNA-strand breaks and are involved in DNA base-excision repair and other repair pathways. Recent studies suggest a link between DNA damage-induced poly(ADP-ribosyl)ation and all tissues at varying levels, with the highest activity in mammalian testis, spleen and thymus. in particular, the poly(ADP-ribosyl)ation is, therefore, pivotal in the maintenance of genomic integrity as well as in the cellular responses to DNA damage. the aim of the present research has been to determine the PARP activity and to study protein expression in the testis of frog, morphological and molecular identified as Pelophylax bergeri, of three different polluted sites and one control site. Antioxidative Glutathione Stransferase activity has been also determined in order to correlate the poly(ADP)ribosylation with the oxidative stress. Moreover the biochemical data have been integrated with morphological evaluation, to assess the health status of the gonadal tissue.

Proteomic Analysis of S-Acylated Proteins in Human B Cells Reveals Palmitoylation of the Immune Regulators CD20 and CD23

S-palmitoylation is a reversible post-translational modification important for controlling the membrane targeting and function of numerous membrane proteins with diverse roles in signalling, scaffolding, and trafficking. We sought to identify novel palmitoylated proteins in B lymphocytes using acyl-biotin exchange chemistry, coupled with differential analysis by liquid-chromatography tandem mas...

Insulin-regulated protein palmitoylation impacts endothelial cell function.

OBJECTIVE Defects in insulin signaling are associated with abnormal endothelial cell function, which occurs commonly in cardiovascular disease. Targets of insulin signaling in endothelial cells are incompletely understood. Protein S-palmitoylation, the reversible modification of proteins by the lipid palmitate, is a post-translational process relevant to cell signaling, but little is known abou...

Detection of Protein Palmitoylation in Cultured Hippocampal Neurons by Immunoprecipitation and Acyl-Biotin Exchange (ABE)

Palmitoylation is a post-translational lipid modification involving the attachment of a 16-carbon saturated fatty acid, palmitate, to cysteine residues of substrate proteins through a labile thioester bond [reviewed in]. Palmitoylation of a substrate protein increases its hydrophobicity, and typically facilitates its trafficking toward cellular membranes. Recent studies have shown palmitoylatio...

Labeling and quantifying sites of protein palmitoylation.

As a reversible posttranslational modification, protein palmitoylation has the potential to regulate the trafficking and function of a variety of proteins. However, the extent, function, and dynamic nature of palmitoylation are poorly resolved because of limitations in assay methods. Here, we introduce methods where hydroxylamine-mediated cleavage of the palmitoyl-thioester bond generates a fre...

Protein Palmitoylation and Its Role in Bacterial and Viral Infections

S-palmitoylation is a reversible, enzymatic posttranslational modification of proteins in which palmitoyl chain is attached to a cysteine residue via a thioester linkage. S-palmitoylation determines the functioning of proteins by affecting their association with membranes, compartmentalization in membrane domains, trafficking, and stability. In this review, we focus on S-palmitoylation of prote...
