چکیده

UNLABELLED PREMISE OF THE STUDY Primula halleri is a high alpine endemic species with a patchy distribution in Central and Eastern European mountains. Little is known about the structure of genetic variation and mating system in populations of this species. • METHODS AND RESULTS We report on the development of 12 novel, polymorphic microsatellite loci for P. halleri. Tests for amplification and polymorphism were performed on 45 individuals sampled from three populations in the Swiss Alps. Interspecific amplification of the loci was evaluated using samples of the closely related species P. farinosa, P. frondosa, and P. scotica. • CONCLUSIONS The results show that the new microsatellite loci will be useful to study the structure of genetic diversity and mating system of P. halleri, and possibly its closely related species.

Characterization of Novel Microsatellite Loci for Primula poissonii (Primulaceae) Using High-Throughput Sequencing Technology.

Primula poissonii (Primulaceae) is a perennial herb, widely distributed in the Hengduan Mountain region of Southwest China. In this study, Roche 454 pyrosequencing was used to isolate microsatellite markers. A total of 4528 unique sequences were identified from 68,070 unique reads. Of these, eighty-seven microsatellite loci were screened for utility using two criteria: successful PCR amplificat...

High-throughput microsatellite marker development for the distylous herb Primula mistassinica (Primulaceae)1

UNLABELLED PREMISE OF THE STUDY Twelve microsatellite markers were developed for Primula mistassinica, a distylous, diploid arctic-alpine plant. The markers will be used to investigate the landscape genetics of a disjunct population on Isle Royale, Michigan, and the phylogeographic patterns of the species. • METHODS AND RESULTS We used Roche/454 high-throughput technology to sequence micro...

Characterization of polymorphic microsatellite markers for Primula sikkimensis (Primulaceae) using a 454 sequencing approach1

PREMISE OF THE STUDY Microsatellite markers from Primula sikkimensis (Primulaceae) were developed for testing deep lineage divergence and speciation events. METHODS AND RESULTS A total of 3112 microsatellites were identified from 61,755 unique reads though 454 pyrosequencing technology. Twenty-nine microsatellite loci were selected for PCR amplification and polymorphic analyses. Among the 29 ...

Development of 13 microsatellite markers in the endangered Sinai primrose (Primula boveana, Primulaceae)1

UNLABELLED PREMISE OF THE STUDY We developed microsatellite markers for the endangered plant Primula boveana, the Sinai primrose, and assessed the cross-transferability of these markers to six related taxa. • METHODS AND RESULTS DNA sequences containing microsatellites were isolated from a microsatellite-enriched library. We obtained successful amplification of 13 microsatellite primer pai...

Development of EST-SSR markers for Primula ovalifolia (Primulaceae) by transcriptome sequencing1

Premise of the study Microsatellite primers were developed for Primula ovalifolia, a member of Primula section Petiolares (Primulaceae), to study the population genetics and species delimitation in this section. Methods and Results A total of 4753 markers were successfully designated from 5139 putative simple sequence repeat loci. We isolated 38 expressed sequence tag-simple sequence repeat m...
