Functionalization of DNA-Dendron Supramolecular Fibers and Application in Regulation of Escherichia coli Association

چکیده
Specific carbohydrate recognition in biology is a dynamic process. Thus, supramolecular multivalent scaffolds with dynamic features have been applied to mimic this process. Herein, we prepared DNA-dendron supramolecular fibers and synthesized carbohydrate-oligonucleotide conjugates (C18mannose). Via DNA hybridization, the C18-mannose could be guided onto the fiber platform and form multiple mannosefunctionalized fibers, which can be utilized to agglutinate E. coli because of high affinity among multivalent mannose ligands and receptors on E. coli. In addition, via chain exchange reaction of DNAs, the E. coli could be dissociated by replacing multivalent mannose ligands with competitive unmodified DNA sequences. The association and dissociation processes of E. coli are confirmed by fluorescent microscope and transmission electron microscope (TEM). These results not only demonstrate the ability of DNA-dendron fibers in reversibly associating E. coli but also illustrate their potential to be an easily modified multivalent supramolecular platform.

Evidence of DNA Transfer through F-pilus Channels during Escherichia coli Conjugation
The mechanism of DNA transfer from Escherichia coli (E. coli) Hfr donor strain AT2453 to recipient strain AB1157 during the conjugation process has been investigated by liquid atomic force microscopy (AFM). With the success of immobilizing both E. coli strains on gelatin-treated glass under aqueous solution, the F-pilus between an E. coli mating pair could be clearly imaged and dissected by an ...

ITALIC! Escherichia coliJF733 is a strain with a long history in research on membrane proteins and processes. However, tracing back the strain development raises some questions concerning the correct genotype of JF733. Here, we present the complete draft genome of ITALIC! E. coliJF733 in order to resolve any remaining uncertainties.

Familial melanoma-astrocytoma syndrome is a tumor predisposition syndrome caused by inactivating germline alteration of the CDKN2A tumor suppressor gene on chromosome 9p21. While some families with germline CDKN2A mutations are prone to development of just melanomas, other families develop both melanomas, astrocytomas, and occasionally other nervous-system neop...
